Panellus serotinus with good nutrition and health value, is one of the favorites ofconsumers and has broad market prospects. In recent years, Panellus serotinus has been firstlycultivated successfully in the northeast of China and bred fine varieties. Because strains willslowly emerge degradation, the ongoing breeding fine varieties is necessary. In this paper,excellent strains were bred by single-single hybridization methods. Through structural botanytest methods, the morphological development of Panellus serotinus were studied, providing thebasis for resource development and utilization of Panellus serotinus.1. Defining the morphological development process of Panellus serotinus(1) Study on morphological characteristics and germination characteristics of the Panellusserotinus basidiosporesThrough microscopic observation, the results showed that the basidiospores were colorless,transparent, containing a nucleus and sausage shape and had smooth surface. Testing thebasidiospores germination in sterile water and liquid PDA culture medium showed that thebasidiospores could not germinate easily in sterile water, but could easily germinate in PDAliquid medium. In PDA liquid medium, the basidiospores expanded within36h, and producedgerm tubes at48h. The germ tubes differentiated into primary hyphae at84h.(2Morphological observation of Panellus serotinus hyphaeObserving the primary hyphae and secondary hyphae of Panellus serotinus under ordinaryoptical microscope and fluorescence microscope, we found that the primary hyphae had lessbranches, no lock-like joint structures, the similar diameter of the main branches and the sidebranches and only one nucleus in a cell; the secondary hyphae had more branches, the largerdiameter of the main branches than the side branches, smaller distance between two adjacentmembranes, lock-like joint structures and two nucleus in a cell.(3) Research on the development of the Panellus serotinus basidiocarpVarious stages of research materials from primordia to mature fruiting bodies were dissectedby paraffin section and freehand section. The results showed that the primordia firstlydifferentiated into stipe followed by the pileus, hymenium finally matured and became maturefruiting bodies. Throughout the development process, hymenium was in an exposed state. So thedevelopment type of the basidiocarp was bare fruit type. The development type of theprimordial was stipe development type and the development type of gills was irregular hymenium type. The gill structure could be divided into hymenia and trama. Hymenium wasmainly composed of rod-like basidia mixed with a small amount of the fusiform cystic body.Each basidium had four sterigmas and one basidiospore was on each sterigma.2. Breeding excellent strains of Panellus serotinus138hybridized combinations were formulated by mon-mon-mating method. Aftercytological identification, physiological characterization identification and biochemicalcharacterization identification,20true hybrid strains were getted. By comparing the myceliumgrowing speed and fruiting characteristics, the fine strains CE89, CE132were screened out. Themycelia of CE89, CE132strains were white and thick dense and growed fast. CE89, CE132strains with short growth duration, high-yield, high biological efficiency, good product traits andstrong resistance had the potential to be developed into new species.
