Momordica seed is the dried ripe seeds of Momordica cochinchinesis (Lour.) spreng. Single and compound recipe of it has been widely used clinically and has demonstrated significant effects, especially in compound recipe of anti-tumor, which take advantage of its effect on detumescence and wound treatment. Although Momordica seed has been used in anti-tumor compound, and occupies a central position in the prescription, its study of material basis is still blank. Therefore looking for the active ingredient of anti-tumor in Momordica seed, and comfirming its material basis, contribute to further evaluating efficacy, and exploring its mechanism. Specific works are summarized as following:The extracting and purification of Momordica developed in Chapter 1, which were divided into two sections. The extraction method of Momordica cochinchinesis was introduced in section 1. While in section 2, semi-preparative high performance liquid chromatography was used to separate the crude extract (F) of Momordica cochinchinesis, and three fractions were separated collected from F, which were named as F1, F2 and F3.The screening of anti-tumor activity in vitro was described in Chapter 2. In vitro, MTT method was used to evaluate the cytotoxicity on multiple cell lines, including Human gastric cancer cell lines SGC-7201, human lung cancer cell lines A549 and H1299, human prostate cancer cell lines PC3 and mice colonic cancer cell lines CT26. F, F1, F2 and F3 showed different levels of anti-proliferative activity against cancer cell and F3 demonstrated most significant effects.The component analysis was described in Chapter 3, which was divided into two sections. In section 1, a reliable and validated high-performance liquid chromatographic assay was established for determination of trypsin inhibitor activity in each part of Momordica cochinchinensis extract, and F1 demonstrated significant activity. While in section 2, LC-(MS)n, HPLC and NMR methods were establish to identify the structure of saponins from Momordica cochinchinensis, which contained in F3. The method of LC-(MS)n was used to infer the structure of saponins through molecular weight and fragment signals. Two simple methods including gas chromatography and high-performance liquid chromatography were established to assay the component of monosaccharide in saponins. NMR including 1H-NMR、BB-DEPT、HMQ、HMBC、1H-1HCOSY and ROESY were used to confirm the structure of saponins from Momordica cochinchinensis.
