| This thesis was mainly concerned the study on the chemical constituents of Melicope patulinervia (Merr.&Chun) Huang, HPLC determination of bioactive component in melicope patulinervia, evaluation of the content of active components and antimicrobial antioxidant activities.(1) Melicope patulinervia (Merr.&Chun) Huang is the rare and endangered plant in the Hainan Province, China. The compounds were isolated by Diaion HP-20, Sephadex LH-20, MCI-GEL and silica gel column chromatographic methods. Physicochemical and modem spectroscopic techniques such as MS, H-NMR, C-NMR and2D-NMR were used to identify the compounds.19constituents were isolated from the aerial part of Melicope patulinervia. Their structures were identified as (E)-Methyl-3-{4-[(E)-3,7-dimethylocta-2,6-dienyloxy]phenyl}prop-2-enoate (MZY-1), Meliternatin (MZY-2),3.5,8-Trimethoxy-6,7:3’.4’-dimethylenedioxflavone (MZY-3), Melibentin (MZY-4),5-hydroxy-3,7,8-trimethoxy-3’,4’-methylenedioxy flavone (MZY-5), Pinostrobin (MZY-6),5-hydroxy-3,6,7,8-tetramethoxy-3’,4’-methylene dioxyflavone (MZY-7),7-(3-Methyl-but-2-enyloxy)-3,5,6,8-tetramethoxy-3’,4’-met hylenedioxyflavones (MZY-8), Evoxanthine (MZY-9),3β-hydroxystigmast-5-en-7-one(MZY-10), Arborinine (MZY-11), Kokusaginine (MZY-12), Phytylhexadecan-oate (MZY-13), Preskimmianine (MZY-14),7-(3-Methy1-but-2-enyloxy)-5-hydroxy-3,6,8-trimethoxy-3’,4’-methylenedioxyflavone (MZY-16), Stigmasterol (MZY-17), β-sitosterol (MZY-18),1-Hydroxy-3-methoxy-10-methyl-10H-acridin-9-one (MZY-19),1,2-dimethylquinolin-4-one (MZY-20). All the compounds were isolated from Melicope patulinervia for the first time, and MZY-16was a new compound.(2) HPLC determination of chemical components in Melicope patulinervia. A HPLC method for simultaneous determination of7kinds of chemical compounds from Melicope patulinervia was taken out.The results shows that leaves of Melicope patulinervia contained a large amout of flavones and alkaloids, while the content of them in leaves was higher than the stems. (3) This part was conceived to evaluate the active ingredients of Melicope patulinervia Huang. The results showed that the70%methanol extracts of the leaves had higher level of total flavonoids, with a content of41.3194±0.071mg·g-1. But it was strange that the contents of total phenol of the extracts of the plant were significantly less than that of total flavonoids.(4) Preliminarily investigate the antimicrobial, antioxidant activities of Melicope patulinervia. The antioxidant activities of the different parts, different solvents extracts of Melicope patulinervia were all weak. The leaf extract showed antimicrobial activity against4of plant pathoqenic funqi species. |
