Objective: To detecting the expression of DIAPH3、E-cadherin、CK andvimentin in colorectal carcinoma and adjacent normal tissue,study their relations withclinic pathological behaviors,such as tumor cell differentiation invasion andmetastasis, Analysis relationship betweeneach proteins, discussed its role in colorectalcancer clinical and meaning finally.Methods: An immunohistochemical and fluorogenic quantitative PCR techniq-ues was used to detect the expression levels of DIAPH3,epithelial markers andmesenchymal markers in colorectal carcinoma, adjacent normal tissue and colorectalcancer cells with different transfer ability.Results:⑴The expressions of DIAPH3in colorectal carcinoma tissues weresignificantly lower than those in the adjacent tissues (P<0.05),and The expressionsof DIAPH3associated with the differentiation degree of colorectal cancer(P<0.05).⑵The expressions of E-cadherin and CK in colorectal carcinoma tissues weresignificantly lower than those in the adjacent tissues (P<0.05),and The expressionsof both associated with the differentiation degree of colorectal cancer(P<0.05),vimentin expression in colorectal cancer was significantly higher than in normaltissue.⑶DIAPH3and E-cadherin expression in low metastatic SW480cells wassignificantly higher in colorectal cancer cells SW620(P <0.05), whereas theexpression of vimentin is less than SW480(P <0.05).Conclusion: DIAPH3expressions were correlated with metastasis of colorectalcarcinoma; colorectal carcinoma increased tumor invasion and metastasis byepithelial-mesenchymal transition mechanism; DIAPH3associated epithelial-mesenchymal transition in colorectal carcinoma,through inhibition of epithelial-mesenchymal transition possibly, thereby inhibiting further development of the tumor.
