Objective:To explore the relationship between Human Annexin v、Apoh proteinexpressi in Term Placenta with HBV DNA load and HBV mother-infant infection.Toexplore HBV intrauterine infection mechanism from the levels of cellular andmolecular.Method: Choose40cases of peripheral blood HBsAg-positive pregnant womenwho were seized and delivery at the Second Affiliated Hospital of NanchangUniversity from july2013to januaryr2014for experimental group. Select20cases ofHBV normal pregnant women for the control group in the same period. Two groupsof pregnant women were have normal liver function. According to the principle ofinformed consent for pregnant women, we collect the elbow vein blood of pregnantwomen before delivery, and collect the cord blood and placenta tissue aseptically inthe delivery time, Then the following studies were performed:1、HBV-DNAin serum was deteeted by PCR-fluorescence probing.2、The HBsAg, HBeAg in the maternal peripheral blood and cord blood weredetected by double antibody sandwich enzyme-linked immunosorbent assay(ELISA).3、The protein distribution of Human Annexin v,Apolipoprotein H in placentasof HBsAg positive pregnant women and normal pregnant women were detected byImmunohistochemistry.Results:1.There were3cases of HBsAg positive in umbilical cord blood of40cases ofHBsAg positive pregnant women, while16cases of HBVDNA positive of30cases ofHBsAg positive pregnant womencases, so the rate of total intrauterine was7.5%(3/40).2.HBV DNA load of the serum HBsAg positive pregnant women is divided into3levels,as follows:<104、104-107、>107copies/ml,There were significant difference indifferent group between intrauterine infection and intrauterineun infected (16.603,P<0.05). 3.There were many brown positive signals distribution in each type of placentacells, The positive signals of human Annexin v、Apoh protein was mostly located introphoblastic cells(TC),villous meseachymal cells(VMC) and villous capillaryendothelial cells(VCEC).Positive signals of the human Annexin v、 Apoh proteinexpression was decreased gradually from the maternal side to fetal side of theplacenta, and the positive signals distributed mostly in the cytoplasm, distributedoccasionally in the nucleus.4.The positive expression rate of human Annexin v、 Apoh protein in TermPlacenta with HBsAg-positive pregnant women in peripheral blood was significantlyhigher than the HBV normal pregnant women with statistical difference between thetwo groups (P<0.05).The difference of human Annexin v、Apoh protein expressionrate between HBVDNA positive and HBVDNA negative in maternal was statisticallysignificant (P<0.05).Expression of human Annexin v and Apoh protein intermplacental have a significant positive correlation (r=0.538,P<0.05).Conclusion:1.The risk factor of HBV mother-infant infection was HBV DNA positive inmaternal serum.which also will be higher if virus content in gravida’serum increase.The risk factor of HBV mother-infant infection also will be higher if virus content ingravida’serum increase.2.The Annexin v and Apoh proteins distributed in various types of cells ofplacental, and the Annexin v and Apoh proteins mainly expressed in the cytoplasm ofthe placental cells, the expression of Annexin v and Apoh proteins are decreasedgradually from the maternal side to the fetal side.3.The expression of human Annexin v and Apoh is obviously up-regulated interm placenta with HBsAg-positive pregnant women,which have positive correlation.Both genes may play an important role in the generation and development of HBVintrauterine infection.
