Objective Examine the expression of IL-27mRNA,IL-27RmRNA,IL-17mRNA,IL-33mRNA in peripheral blood mononuclear cells (PBMCs) and the levels ofIL-27,IL-17in serum of the patients with rheumatoid arthritis.To explor thecorrelationship among IL-27,IL-27R,IL-17,IL-33and disease activity in RApatients.Methods The IL-27、IL-17levels in serum of40RA patients and13healthycontrols(HC)were detected by ELISA.The Expression of IL-27mRNA,IL-27RmRNA,IL-17mRNA,IL-33mRNA in peripheral blood mononuclear cells(PBMCs) of30RA patients and10healthy controls were determined by real-timequality PCR(RT-PCR)Results1The IL-27, IL-17levels in serum of RA patients is much higher than the healthycontrols[(49.21±6.56)pg/ml,(112.84±43.96) pg/ml],and there were significantdifference among the high activity,moderate activity,low activitygroups[(171.82±27.70)pg/ml vs (128.39±11.34)pg/ml vs (81.32±5.57)pg/ml];[(566.95±40.74) pg/ml vs (382.28±60.84)pg/ml vs (228.43±47.99)pg/ml];2IL-27,IL-17levels were correlated positively with DAS28,ESR,CRP,SJC,TJC,self assessment,CCP-antibody(all the P<0.01)3The levels of IL-27correlated positively with IL-17((r=0.954,P<0.01)in serumof RA patients.4The expression of IL-27mRNA in peripheral blood mononuclear cells wassignificantly up-regulated in RA patients among high activity、 moderateactivity、low activity groups and HC[(5.142±1.178) vs (3.028±0.897)vs(2.218±0.387)vs(1.137±0.437), the all P<0.01but P=0.113between moderate and low activity groups],and the expressions of IL-27mRNA werecorrelated positively with disease activity.5The expression of IL-27RmRNA, IL-33mRNA, IL-17mRNA in peripheralblood mononuclear cells was significantly up-regulated in RA patients amonghigh activity,moderate activity,low activity groups and HC [(5.762±1.454) vs(3.254±0.776) vs(2.043±0.474)vs(1.100±0.309)];[(2.009±0.156) vs(1.719±0.216)vs(1.219±0.078)vs(1.017±0.142)];[(2.101±0.233) vs(1.465±0.156)vs(1.207±0.130)vs(1.023±0.164)],the all P<0.016The expression of IL-27mRNA in peripheral blood mononuclear cells wascorrelated positively with DAS28score,ESR,CRP,SJC,TJC,selfassessment,CCP-antibody and IL-27RmRNA,IL-33mRNA,IL-17mRNA arealso correlated positively with these items(all the P<0.01),and there were noobvious relations with course of the disease and RF7The expression of IL-27mRNA、IL-27RmRNA、IL-33mRNA、IL-17mRNAwere correlated positively with each other in RA patients8The levels of IL-27was correlated positively with the expression of IL-27mRNA in peripheral blood mononuclear cells in RA patients(n=30,r=0.848,P<0.01),and it is same to IL-17ConclusionThe expression of IL-27mRNA, IL-27RmRNA,IL-33mRNA,IL-17mRNA wereup-regulated and they were correlated positively with each other and RAdisease activity.In addition the levels of IL-27and IL-17were higher in RApatients and they were all correlated positively with RA disease activity. IL-27level was correlated positively with IL-17level, suggesting IL-27,IL-17andIL-33may interact with each other and they maybe play an important role in thepathogenesis and disease activity of RA...
