Objective To investigate the influence of Apolipoprotein A-I mimetic peptide D-4Fon proliferation and migration of vascular smooth muscle cells(VSMCs)Methods Rat aortic VSMCs were isolated and cultured under standard conditions.VSMCs were treated with D-4F or ox-LDL at varying concentrations for differenttime intervals and used for several analyses. CCK-8kits were used to analyze cellproliferation. Cell migration was determined with a transwell chamber and ascratch-wound assay. Intracellular reactive oxygen species (ROS) production wasassessed based on ROS-mediated DCF fluorescence.Results Ox-LDL promoted VSMCs proliferation and migration. However, D-4Fcould efficiently inhibit VSMCs proliferation and migration induced by ox-LDL.Forthermore, we observed that ox-LDL remarkably enhanced ROS generation. D-4Fcould efficiently inhibit ROS production triggered by ox-LDL and subsequentlyblocked the proliferative and promigrative effects of ox-LDL in VSMCs.Conclusion D-4F could inhibit ROS production triggered by ox-LDL andsubsequently blocked the proliferative and promigrative effects of ox-LDL in VSMCs,which could be closely related to its anti-atherogenic roles.
