Aim:To explore the effect of recombinant human growth hormone on intestinal mucosa of rats with obstructive jaundice and tight junction protein claudin-1and mitogen activated protein kinase2.Methods:Eighty adult Wistar Rats(any male and female) were randomly divided into4groups:Ⅰ, Sham Operation (SO, n=12); Ⅱ, Obstructive Jaundice (OJ, n=28); Ⅲ, Recombinant Human Growth Hormone (rhGH, n=28);IV, Sham Operation with Recombinant Human Growth Hormone(GSO,n=12). In OJ and rhGH group, rats were randomly assigned into7days (n=14) and14days (n=14) group. In SO group,rats were randomly assigned into7days (n=14) and14days (n=14) group.The production model jaundice rat obstruction. After operation rhGH group and GSO group underwent double hind leg turns subcutaneous injection of recombianant human growth hormone(0.75iu/kg/d).After7days,14days collecting rat blood,retrieve the terminal ileum intestinal tissue rat blood liver function tests. To observe the change of mucosa pathological morphology of intestinal tissue of claudin-1protein and MEK2content closely connected.Result:(1) TB、DB、ALT in the groups of OJ and rhGH were average higher than those in the groups of SO (P<0.05).(2) RhGH-7day group than in the GSO-7day group of intestinal mucosal injury severity(P<0.05); RhGH-7day group than in the OJ-7day group of intestinal mucosal injury degree of light(P<0.05); RhGH-14day group than in the OJ-14day group of intestinal mucosal injury degree day light(P<0.01)(3) The expression level of rhGH group,OJ group of intestinal mucosal tight junction protein claudin-1is lower than that in SO group (P<0.01);expression level in rhGH group than OJ group,intestinal tight junction protein claudin-1(P<0.05).(4) The expression level of mucosa, intestinal rhGH MEK2OJ group than in SO group (P<0.01);the expression level of rhGH group than in OJ group decreased intestinal mucosal MEK2(P<0.05).Conclusion:(1) Recombinant human growth hormone could up regulate the expression of intestinal mucosa in rats with obstructive jaundice Claudin-1. (2) Recombinant human growth hormone on the regulation of MEK2/MAPK signaling pathway to improve the blocking barrier function of intestinal mucosa in rats with obstructive jaundice.
