Objective:To investigate the effect of recombinant growth hormone on tight junction associated protin zo-1and phosphorylation myosin light chain of intestinal mucosa in rats with obstructive jaundice.Methods:Seventy adult Wastar Rats(any male and female) were randomly divided into3groups:I. Sham Operation (SO, n=10); II, Obstructive Jaundice (OJ, n=25); III, Recombinant Human Growth Hormone (rhGH, n=25);IV, Sham Operation with Recombinant Human Growth Hormone(GOJ,n=10). In OJ and rhGH group, rats were randomly assigned into7days (n=12) and14days (n=13) group. In SO group,rats were randomly assigned into7days (n=10) and14days (n=10) group.Group I and groupIV:20rats underwent laparotomy, but common bile duct (CBD) was not ligated. Group II and III:50rats underwent laparotomy, and CBD was ligated. In the group of rhGH and GOJ, Rats were undertaken subcutaneous injected of rhGH on posterior inside limb each day with dosage0.75iu/kg/day. On postoperative7days and14days, ligate the terminal segments of ileum and inject fluorescein-isothiocyanate-dextran (FITC-Dextran) in these segments of ileum.then the blood of portal vein、 the blood in herat and the terminal segments of ileum were harvested. Ileum stained with hematoxylin and eosin was examined histologically and morphometrically. To measure the TB、DB、 ALT with heart blood.The intestinal permeability of rats was measureed by FITC-dextran with portal vein blood.The changes of tight junction associated protein ZO-1and phosphorylated MLC (PMLC) in intestinal epithelia of the rats were analyzed by inmmunohistochemistry with the terminal segments of ileum.Result:(1) TB、 DB、 ALT in the groups of OJ and rhGH were average higher than those in the groups of SO (P<0.05); TB、 DB、 ALT in the groups of OJ and rhGH were gradually increased with the time going (P>0.05); There were not significant difference between the groups of OJ and rhGH on TB、 DB、 ALT,(P>0.05).(2) The ileal mucosa of group OJ and rhGH were acutely injuried than group SO (P<0.01).While the time going the degree of ileal mucosa injuried in group OJ was increased(P<0.05). The degree of ileal mucosa injuried in group rhGH-7days was lower than in group OJ-7days(P<0.05); The degree of ileal mucosa injuried in group rhGH-14days was lower than in group OJ-14days(P<0.01). The degree of ileal mucosa injuried in group rhGH-14days was lower than in group rhGH-7days, but there were not significant difference (P>0.05).(3) The absorption strenth of fluorescein-isothiocyanate-dextran(FITC-dextran) in the groups of OJ and rhGH were higher than those in the groups of SO (P<0.05); The absorption strenth were gradually increased with the time going (P<0.05); The absorption strenth of FITC-dextran in the group of rhGH were less than those in the group of OJ(P<0.05).(4) The expressions of tight junction associated protein (TJP)ZO-1in the groups of OJ and rhGH were less than those in the group of SO (P<0.01); The expressions of TJP ZO-1in the groups of rhGH were higher than in the group of OJ (P<0.01); The expressions of TJP ZO-1in the group of OJ-14days was less than those in the group of OJ-7days (P<0.05); There were not significant difference between the groups of rhGH-14days and rhGH-7days on the expressions of TJPZO-1(P>0.05).(5) The expressions of phosphorylation myosinlight chain(PMLC) in the groups of OJ and rhGH were less than those in the group of SO (P<0.01); The expressions of PMLC in the groups of rhGH were higher than in the group of OJ (P<0.01); The expressions of PMLC in the group of OJ-14days was less than those in the group of OJ-7days (P<0.05); There were not significant difference between the groups of rhGH-14days and rhGH-7days on the expressions of PMLC (P>0.05).Conclusion:(1) Recombinant Human growth hormone can increase the expression of TJP ZO-1and reduced the expression of PLMC of intestine mucosal in rats with obstructive jaundice.
