Object iveTo comprehensively analyse and compare the main component of the pieces of Ling Nan characteristic for Cornu Cervi Pantotrichum (LNCD) and its Chinese Pharmacopoeia product, for analysing the feature of main components in LNCD and interpreting the rationality of the traditional processing and features of LNCD. In order to standardize and promote the processing of LNCD, we used single factor test to optimize the best processing technology in key, used new technology of drying at room temperature to optimize the best drying progress. The quality of Ling Nan traditional Cornu Cervi Pantotrichum prepared for decoction(LNCD) were studied in this paper, which was used to builded the quality standard draft and drafting instruction. The mutual mode of HPLC fingerprints was builded for comparing the similarity of Cornu Cervi Pantotrichum from different process, Which are benefit for us to discover the cooking significance of LNCD.Methods1a study on the methods of main component contentUsing water soluble protein (WSP), total phospholipids (TP), total polysaccharides anthrone (TPA), total free amino acid(TFAA) and dissolution rate of water-soluble extract (DRWE)as the evaluating standard to analyse TLNP and its Chinese Pharmacopoeia product. And those contents were respectively determinate by the methods of Coomassie blue colorimetric, molybdenum blue colorimetric, sulfuric acid-Determination and ninhydrin color and determination of boil water-soluble extract.2a study on the processing technology of TNCDWe used the degree of embellish and softening as index, used single factor test to optimize the best processing technology in experiment. New normal temperature drying technology have been introduced, and we used the content of alcohol-soluble extract, total free amino acid(TFAA) and identification in appearances as the test index. At the same time, drying temperature, drying time, and material density were considered as examine factors to optimize the drying process test.3a study of qualityRefer to "Chinese pharmacopoeia"2010edition, we determined moisture content, alcohol-soluble extract content, total ash content and established the methods of appearances identification, microscopic identification, TLC identification, content determination. According to the result of the research from10batch LNCD determination, we builded the quality standard draft and drafting instruction.4a study of HPLC fingerprintUsing HPLC-PDA as full wavelength scanning method of hydrochloric acid extract of LNCD to ascertained the baseline stability and provide detection wavelength of information. And using uracil as a control article to established fingerprint and obtained the Common Peaks in the HPLC fingerprints (HPLC-FPS) of LNCD, using "traditional Chinese medicine fingerprint atlas of similarity evaluation system2004A version" to analyzed the Common Peaks similarity.Results1a study on the methods of main component contentThe results show that TPA and TFAA of TNCD had a positive correlation with TNCD grades, but WSP and TP without this characteristic. The content of evaluating standard in TNCD were higher than that in its Chinese Pharmacopoeia product except free amino acid, respectively was0.82%,0.097%,0.23%, and the difference between those in WSP and TFAA possessed statistically significant. The dissolution rate of water-soluble extract of TNCD was significantly higher than that in its Chinese Pharmacopoeia product, which dissolution percentage of quality was94.2%at the time of10min, under the same condition the percentage of its Chinese Pharmacopoeia product was60.6%.2a study on the processing technology of TNCDBased on the test results and the characteristics of the Ling Nan weather, Specific research content as follow:the liquid ratios of rice wine volume is1:3; the segments of tender Cornu Cervi Pantotrichum are chopped3h, and the harder ones are chopped5h in spring and summer. The segments of tender Cornu Cervi Pantotrichum are chopped4h, and the harder ones are chopped6h in fall and winter. Then stuffy embellish2d; the segments of tender Cornu Cervi Pantotrichum are steamed2h, and the harder ones are steamed3h in the first time. At the second and the third time, steam them10mins. According to the experimental results, the production efficiency and cost, we finally find that the best technology is drying4h at35℃when the drying density is9g.3a study of qualityDetermination methods of appearances identification, microscopic identification, TLC identification, content determination were obtained in this article. Besides, by analyzing the determination result, we ascertained moisture content, alcohol-soluble extract content, TFAA content determination as detection project.4a study of HPLC fingerprintThe HPLC fingerprints (HPLC-FPS) of10batches LNCD were built in this article, which included20Common Peaks. The similar degrees of HPLC fingerprints of Cornu Cervi Pantotrichum from different process and qualifications were compared, which indicated that the LNCD from different qualifications was little different, and the LNCD from different process was more different, the component species among those were basic equal.conclusion1a study on the methods of main component contentTFAA content basically reflects the scientific rational of traditional quality level classification in LNCD and provides reliable quality evaluating method for LNCD. The different of main components between LNCD and and its Chinese Pharmacopoeia product is consistent with the function of steaming process. LNCD’s characteristic enables the active ingredients to Maximum used, its Chinese Pharmacopoeia product does not have this feature. 2a study on the processing technology of TNCDFurther details in processing technology of LNCD is obtained in this article, and new technologies drying at rooms temperature is introduce to promotes the efficiency in production and the quality in product, which will be benefit for providing basic methods and foundation for LNCD.3a study of qualityThe quality standard of LNCD is draft as follow:LNCD ought to conforms the demand of identification in appearances, microscopy, TCL, and the moisture content cannot exceed13.0%, the content of alcohol-soluble extract cannot lower than5.0%, the content of total free amino acid cannot lower than0.90%.4a study of HPLC fingerprintThe determination method of HPLC fingerprint is stable, reliable, and which provides basis in evaluate the quality of Cornu Cervi Pantotrichum prepared for decoction.
