Objectives: Renin-angiotensin system (RAS) plays a very important role in theregulation of blood pressure. We hypothesize that the genetic polymorphisms in RASmight be associated with elevated blood pressure. We aimed to investigate theassociations of angiotensinogen gene (AGT)-rs5051, rs699, rs5046polymorphisms,and angiotensinâ…¡type1receptor gene (AT1R)-rs5186polymorphisms with bloodpressure in Chinese.Methods: We invited all unrelated employees and retired workers to participate in ourstudy from May to July,2010. After excluding those with missing demographicinformation or those without DNA,1464participants were included in the presentanalyses. We administered a standardized questionnaire to collect information onsmoking habits, alcohol consumption, medical history and medications. We alsoperformed body height, body weight, and waist and hip circumferences measurements.Overnight fasting blood was collected to perform biochemical measurements. Geneticpolymorphisms were genotyped using SNaPshot methods.Results:The genotype frequency of AGT-rs699was AA4.06%, GA34.40%, and GG61.54%, respectively. The genotype frequency of AGT-rs5046was AA2.78%, GA25.43%, and GG71.79%, respectively. The genotype frequency of AGT-rs5051wasCC4.06%, CT34.62%, and TT61.32%, respectively. The genotype frequency ofAT1R-rs5186was AA90.5%, CA9.4%, and CC0.1%, respectively.There was no significant difference in the distributions of genotypes andalleles between normotensive participants and hypertensive patients (P>0.05) inAGT-rs5051, rs5046, rs699and AT1R-rs5186. Furthermore, there was no significant difference in systolic and diastolic blood pressure and pulse pressure betweengenotypes both before and after adjusted for covariate (P>0.05).There was a significant interaction between AT1R-rs5186(A1166C) andbody mass index to regulate systolic (Pint=0.004) and diastolic blood pressure (Pint=0.04) and pulse pressure (Pint=0.02). Indeed, only in participants in the highestquartile of body mass index, C allele carrier had a significantly higher systolic (P=0.03) and diastolic blood pressure (P=0.05) and pulse pressure (P=0.03) than AAhomozygotes.Conclusions:1. AGT-rs699, rs5051, and rs5046polymorphisms were not significantlyassociated with blood pressure in Chinese in Dali.2. AT1R-rs5186polymorphism was not associated with blood pressure, butrs5186interacted with body mass index synergistically to regulate blood pressure inChinese.
