Experimental Research To Yishenjiedutongluo Capsule On The Level Of CTGF、FN Of Diabetic Nephropathy Rats

Objective: To induce the SD rat model of diabetes mellitus with STZ,and treatlosartan potassium as the positive control,to confirm the impact of theYishenJieduTongluo capsule on the expression of Connective Tissue Growth Factorand fibronectin,and the impact of the capsule on the renal protection of diabetesmellitus rats. To explore the relation of the "toxin impairing renal collaterals" theoryand the pathogenesis of diabetic nephropathy.Methods:60male SD rats,10rats were treat as lank group, and the oher50ratswere treated with intraperitoneal injection of STZ to induce diabetes mellitus modeland then randomly divided into5groups: model group, normal control group,Yishejieduntongluo capsule high dose group, middle dose group and low dosegroup,and each group has10rats. All groups were given suspension gavage andcontinuous adiabetes mellitusinistration for12weeks, then execution of weight, bloodglucose, blood lipid, renal function, urinary protein, and kidney hypertrophy indexindicators,then observed the pathological changes of kidneys with optical microscope,explored the expression of CTGF and FN by immunohistochemical staining.Results:1.the Yishejieduntongluo capsule can inhibit the decline of weight ofdiabetes mellitus rats, reduce the increase of the urine volume(P<0.01), and improvethe general conditions of diabetes mellitus rats;2. the Yishejieduntongluo capsule cansignificantly lower the level of the blood glucose, blood lipid of diabetes mellitusrats(P<0.01);3. the Yishejieduntongluo capsule has the function to lower the kidneyhypertrophy index(P<0.01), and reduce the creatinine and urea nitrogen, inhibit theendogenous creatinine clearance, which is superior to losartan potassium(P<0.05,P<0.01);4.the Yishejieduntongluo capsule can reduce the24h urinay proteinexcretion,and its efficacy is superior to losartan potassium(P<0.01);5.treatmentgroup has better condition than that of the model group in renal pathologicalchanges after the observation of the optical microscope, and the pathological change of the high dose group was better than other groups;6.the Yishejieduntongluo capsulecan reduce the expression of the CTGF and FN in renal tissue(P<0.01), and theeffecacy is superior to losartan potassium(P<0.01).Conclusion: The Yishejieduntongluo capsule can sinificantly inhibit thedevelopment of diabetic nephropathy induced by STZ and improve the general stateand lower the level of the blood glucose, blood lipid of diabetes mellitus rats,andinhibit the expression of CTGF and FN.The capsule has the significant protection andimprovement of the structure and function of the kidneys of diabetes mellitus rats,andthe mechanism may be related to the inhibition of the expression of CTGF and delayof the renal fibrosis.
