【Objective】This study was to investigate the expression level of USP22protein of cancerstem cell markers in laryngeal squamous cell carcinoma (LSCC) samples and toevaluate the correlation between the expression of USP22protein andclinicopathologic factors,such as clinical stage differentiation,lymph node metastasisand prognosis of LSCC.【Methods】The expression of USP22protein was detected by immunohistochemistry in64cases of carcinoma tissue(LSCC) and26cases of squamous epithelium besidecarcinoma tissues(Control). Collect clinical and prognostic information of64patientswith laryngeal squamous carcinoma in detail.We analyzed the correlation between theexpression of USP22protein and various clinicopathologic factors including survivalstatus through Kaplan-Meier method and the Log-rank test.【Results】1. The expression level of USP22protein in LSCC and control were57.81%and19.23%, the difference was statistically significant(P=0.001).2. The expression level of USP22protein in laryngeal squamous carcinoma clinicalstaging Ⅰ+Ⅱstage and Ⅲ+Ⅵstage high expression rate is41.4%and71.4%respectively, the difference was statistically significant (2=5.871, P=0.015); Theexpression level of USP22protein in laryngeal squamous carcinoma TNM stage T1+T2stage and T3+T4stage high expression rate is44.4%and75.0%respectively, the difference was statistically significant (2=6.029, P=0.014),The expression level ofUSP22protein in laryngeal squamous carcinoma without lymph node metastasis andlymph node metastasis high expression rate is48.9%and78.9%respectively, thedifference was statistically significant (2=4.949, P=0.026).Clinicopathologicalanalysis showed that USP22expression level had a relationship withdifferetiation,clinical stage (p=0.016) T stage (p=0.014), and lymph node metastasis(p=0.026), but not with gender,age smoking and differentiation.3.64cases of laryngeal squamous carcinoma patients’ overall survival rate was73.1%, The high expression level of USP22protein and the low expression level ofUSP22protein in laryngeal squamous carcinoma on the5-year survival rate is62.1%and88.06%respectively, the difference was statistically significant (P <0.05); TheSupraglottic type laryngeal squamous cell carcinoma,the glottis type laryngealsquamous cell carcinoma, the subglottic laryngeal squamous cell carcinoma on the5-year survival rate is64.1%,81.1%and64.1%respectively; Different types of5-year survival rate from high to low order is the the glotti type,Supraglottictype,subglottic type.4.In univariate analysis, the survival was related to clinical stage (P=0.006), TNMstage (P=0.043), whether lymph node metastasis (P <0.001) and differentiation(P=0.001)the difference was statistically significant (P <0.05); while age, sex,smoking, were not correlated to prognosis, There was no statistically significantdifference (P>0.05).【Conlusion】1.The expression of USP22was significantly higher in LSCC than in control group,which overexpression may involve in the development process of laryngeal squamouscarcinoma. 2. The high expression level of USP22protein and clinical staging of laryngealsquamous cell carcinomas, infiltrating depth and lymph node metastasis related. Withthe increase of laryngeal squamous carcinoma infiltration depth, USP22proteinexpression is higher, USP22unusually high protein expression may be involved inthe occurrence, invasion and metastasis process of laryngeal squamous carcinoma.3. Survival analysis was shown that patients with high USP22expression hadsignificantly poorer outcome compared with patients with low expression of USP22;Different types of5-year survival rate from high to low order is the the glottitype,supraglottic type, subglottic type.4. The high expression level of USP22protein and the low differentiation, clinicalstage and lymph node metastasis of laryngeal squamous cell carcinomas, are theimportant factors influencing the prognosis of laryngeal squamous cellcarcinomas,the detection of USP22may be serve as a new candidate molecularmarker and therapeutic targets of LSCC patients.
