| Objective:By perfusing mice with contionuous pungent food to induct allergic reaction model, the author observes responses of preservative(that is ethylparaben) and hair dye (that is p-Phenylenediamine) to stimulation of the skin.Meanwhile we apply the method of RT-PCR to test the expressions of Chemokine Receptors (CXCR3,CCR5and CCR3) in skin and blood and to probe function and mechanism of Chemokine Receptor in allergic dermatitis, so as to supply experimental bases for the prevention and treatment of related diseases.Methods:1. We randomly divide140Kumming female mice which are weihgted at18to20gram into seven groups, namely the blank group, the ethylparaben group,the pungent ethylparaben group, the p-Phenylenediamine group,the pungent p-Phenylenediamine group, the mixed group, and the pungent mixed group.Mice are perfused with the mixture which includes chili solution and Chinese spirits10ml/(kg.d) and some mice are given physiological saline once a day.All these mice are needed seven days to be assessed on the basis of their signs. We give the animals an abdominal injection of reagent solution of two reagents after Spicy model is successfully made. Then after seven days we apply respectively reagent to the back of the animals and after anther forty-eight hours, based on the cutaneous reactions we evaluate the score according to Cosmetic Health Norms.2.We take the mice skin of2cm*2cm, of which is partially embedded in paraffin to have pathologic analysis and the rest is to be extracted to RNA.3.We collect blood of one point five milliliter in the method of eyeball blood,using lymphocytes separation medium to separate mononuclear cells and extract RNA.4.We test the expressions of skin and CXCR3, CCR5and CCR3in PBMC(Peripheral Blood Mononuclear Cell) by means of RT-PCR.Results:1. Resutls in state of animal stimulation:The pungent group have striking Irritation symptoms and obvious differences by statistics (P<0.01).On Skin allergic reaction and Pathological score, the scores of the pungent group rise, compared with the non-pungent groups and have statistical meaning (P<0.01); The scores of the mixed group go up, compared with the single group and have statistical meaning (P<0.01); The pungent mixed group compared with the pungent single group have stiking differences (P<0.01) by statistics.2.Results in skin:Compared with non-pungent group,the scores of the pungent group CXCR3and CCR5rise and have statistical meaning (P<0.01); the scores of the mixed group CXCR3ã€CCR5go up, compared with the single group CXCR3and CCR5and have statistical meaning; The experimental group CCR3has no significant change and statistical meaning (P>0.05).3. results in blood:Compared with non-pungent group,the scores of the pungent group CXCR3and CCR5rise and have statistical meaning (P<0.01); the scores of the mixed group CXCR3ã€CCR5go up, compared with the single group CXCR3and CCR5and have statistical meaning; The experimental group CCR3has no significant change and statistical meaning (P>0.05).Conclusion:The pungent food stimulus aggravates the allergic contact dermatitis caused by preservative and hair dye. Moreover the stimulation displays more obvious when preservative and hair dye are mixed.The Thl Chemokine receptors in allergic contact dermatitis rise evidently in spicy state. |
