Objective:To observe the change of HPA axis of depression models on rats and themechanism of the treatment by Chaihu Shugan Powder on it to explore thepathogenesis of depression and the syndrome of liver depression and the change ofHPA axis,further reveal the mechanism of syndrome and treatment of this disease.Method:1, Grouping: Healthy45male SD rats were randomly divided into3groups1:normal control group,depression model group and Chaihu Shugan Powdertreatment group,15rats in every group.2, Modeling and administration:Besides thenormal group, the rats in the other2groups were fed in the manner of chronicunpredictable mild stimulation for21days.Chaihu Shugan Powder were given tothe rats of Chaihu Shugan Powder treatment group during the course of modelreplication.The rats of normal control group and depression model group weregiven normal saline in the same volume.Measured the body weight and the OFTbehavior study evaluation every week during the course of model replication.3,Pathology observation:To observe the pathological changes in the hippocampus,hypothalamus,pituitary and adrenal cortex tissue morphology of opticalmicroscope.4,Target observation:western blot had been used to determine theexpression of GR and MR in hippocampus, CRH in hypothalamus;Elisa had beenused to determine the plasma ACTH and the serum CORT level.Results:1,3weeks after the the molding,the majority of depression model groupappeared apathetic, limbs, reduced activity, lack of interest, weight loss, OFTbehavioral horizontal sports and vertical sports were significantly lower than thosein normal control group(P <0.05), indicating success of depression modelrats.Compared with the depression model group,Chaihu Shgan Power increased thebody weigh and the scores of OFT.2, Pathology observation: Compared withnormal control group,the rats of depression model group had cell morphologydamage of hippocampus,hypothalamus,pituitary and adrenal.Compared withdepression model group,in Chaihu Shugan Powder group,cell morphology damageof those tissue were slightly.3,Target observation:Compared with normal controlgroup:The expression of GR in hippocampus was significantly decreased (P <0.05);the expression of CRH in hypothalamus was significantly increased (P<0.05), plasma ACTH and serum CORT content significantlyincreased(P<0.05);compared with the depression model group:The expression ofGR in hippocampus was significantly increased (P <0.05);the expression of CRHin hypothalamus was significantly decreased (P <0.05), plasma ACTH and serumCORT content significantly decreased(P<0.05);There were no significantdifference in the expression of MR in hippocampus between each group.Conclusion:1,The rats of depression model group had cell morphology damage ofhippocampus,hypothalamus,pituitary and adrenal,Chaihu Shugan Powder canimproved or basically recover the cell morphology of those tissue.2,The rats ofdepression model has hyperfunction of HPA axis.Prompting that the developmentof depression and hyperfunction of HPA axis are closely related. Chaihu ShuganPowder can improve the depression symptoms of experimentalanimals.3,Antidepression function of Chaihu Shugan Powder may be related to itsincrease the expression of GR in hippocampus,decreased expression of CRH inhypothalamus, plasma ACTH and serum CORT content and,reduce the damage ofhippocampus,inhibit the hyperfunction of HPA axis.5,Syndrome of liver depressionis the major syndromes of depression,Chaihu Shugan powder to the depression ratmodel intervention is effective,and then verify the syndrome of liver depressionand hyperfunction of HPA axis are related.
