| Objective: Discussion Qiliqiangxin capsule on heart failure after myocardialinfarction in rats calcium/calmodulin-dependent protein kinase Ⅱ, and exploreQiliqiangxin capsule treatment of chronic congestive heart failure mechanism.Methods: Health Sprague Dawley (SD) male rats weighing220±30g, the leftanterior descending coronary artery ligated rats, acute myocardial infarction (AMI)model of heart failure, a total of20rats were randomly divided into Qiliqiangxincapsule and saline groups; another left thoracotomy in the left anterior descendingcoronary artery in rats playing a loose knot, the establishment of the sham group10.Qiliqiangxin capsule group, after giving the powder (0.6g/kg d)3ml of normalsaline administered treatment, saline group and the sham group received the samevolume of saline for4weeks prior to treatment, after the rats were detected byultrasound of the heart and blood plasma BNP, after4weeks of treatment weresacrificed to detect myocardial CaMK Ⅱ content and activity in rats.Results:1.modeling results: modeling process due to infection, arrhythmia and otherreasons32rats were killed after the successful model of heart failure aftermyocardial infarction in20rats were randomly divided into Qiliqiangxin group andthe control group (each10) and sham group (10) for comparison.2. Echocardiography results showed: no significant difference (P>0.05) betweencardiac ultrasound the index and BNP among the three groups before surgery. After4weeks, the left ventricular end-diastolic diameter, left ventricular end-systolicdiameter, left ventricular end-systolic volume, left ventricular end-diastolic volumein the control group and Qiliqiangxin group, were large compared with the shamgroup, the difference was statistically significant (P<0.05); The control group leftventricular end-diastolic and left ventricular end-systolic diameter is larger thanQiliqiangxin group, the difference was statistically significant (P<0.05); In thecontrol group compared Qiliqiangxin left ventricular end-systolic volume and leftventricular end-diastolic volume showed no statistically significant (P>0.05);ventricular fractional shortening and left ventricular ejection fraction in the controlgroup and Qiliqiangxin group were lower than the sham group, the control group also below Qili strong heart group, the difference was statistically significant (P<0.05). Control group and Qiliqiangxin group plasma BNP levels were significantlyhigher than the sham group, while the control group was higher than that of plasmaBNP Qiliqiangxin group, the difference was statistically significant (P<0.05).3. Western blot showed: Expression of both CaMKⅡprotein3groups protein shamgroup of CaMKⅡδ(A-17) were lower than Qiliqiangxin group and the controlgroup, the expression Qiliqiangxin group CaMKⅡδ(A-17) than the control grouplow sham group CaMKⅡ(Thr286) protein expression were lower than the other twogroups, the expression Qiliqiangxin group CaMKⅡ(Thr286) compared with thecontrol group. Control group CaMKⅡδ(A-17) bands than the other two groupswere wide, Qiliqiangxin group CaMKⅡδ(A-17) bands compared with the shamgroup wide group CaMKⅡ(Thr286) bands than the other two groups were wide,Qiliqiangxin group CaMKⅡ(Thr286) band width than the sham group. CaMKⅡprotein expression results showed: Sham group cardiomyocytes CaMKⅡδ(A17)protein expression were lower than Qiliqiangxin group and the control groupbetween the two groups was statistically significant (P<0.05); Qiliqiangxinmyocardial fine CaMKⅡδ(A17) protein expression levels than the control group,the two groups was statistically significant (P<0.05); sham group cardiomyocytesCaMK Ⅱ (Thr286) protein expression were lower than Qiliqiangxin group andcontrol group, a statistically between the two groups significance (P<0.05); proteinexpression of CaMKⅡ (Thr286) Qiliqiangxin myocardial cells than the controlgroup, the two groups was statistically significant (P<0.05).Conclusion:1.Qiliqiangxin capsules rats with heart failure after myocardialinfarction can improve relevant indicators echocardiography: left ventricularend-diastolic diameter, left ventricular end-systolic diameter, left ventricular ejectionfraction, left ventricular shortening fraction and improve plasma BNP levels.2.Qiliqiangxin capsules can reduce the amount of protein expression in heart failureCaMK Ⅱ necrotic myocardium after myocardial infarction in rats, can significantlyimprove cardiac function in heart failure after myocardial infarction in rats. |
