The Experimental Research About The Effect Of "Tong Jing Huo Li Tang" On The Cytokine TNF And IL-12of Mice Airbag Synovitis Model

Objective: This research, under the guidance of TCM Theory,used ourhospital’s traditional prescription “Tong Jing Huo li Tang” to treat the back gasbag synovitis model of mice, and observe the effect of thismedicine on cytokine TNF and IL-12on in lavage of mice back gasbag. Through this research, we can make a preliminary discussion about the mec hanism of “Tong Ji ng Huo l i Tang” in curing synovitis, geta further understanding about the mechanism of anti-infla mmatory reaction, and make a prelimina ry analysis for its curative effect. This research can provide a scientific basis for its clinical application.Methods: The experiment used60healthy female Kun Ming mice as the research objects. After unifor m number, these mice,according to random number table, are divided into A: Normal control group B:Blankmodel c ontrol group, C: Tong Jing Huo li Tang group D:Hua Mo Yan Ke Li group. When the experime nt began, we injected5ml air into the back subcutaneously of B、C、D groups on the first、the third、and the sixth days,and kee p the back sacs of the air expansion for7days. On the first day of t his experiment, irr igate the mice of B group by usi ng “Tong Jing Huo li Tang”, irrigate the mice of C group by using “Hua Mo Yan Ke Li”(2.73g·kg－１), irrigate the mice of A and B group by usi ng an equal amount of saline as contrast. Each group irrigated2ti mes a day, seven days in a row. ON the seventh dayof this experiment,2hours after the last irrigate, injecte the mice of B、 C、 D groups on their back airbag by c arageen glue (Car)25mg/kg in order to induced inflammation (A group given an equal a mountof solvent). After cause infla mmation6hours, kill the mice, and injecte4ml HBSS lavage to back airbags, collect lavage fluid. Take a s mall a mount of la vage for white blood cell count, and the rest lavagecentrifugal.(4℃,1800r·mi n-1,10min). Collect clear fluid, detectTFN and IL–12with double antibody sandwich enzyme-linked i mmune experiment (ELISA). USE SPSS17.0statistical software analysis totest results data.Results:(1)At the start of the experiment, the mice’s spirit and the gl-osss’s col our were not equal to the nor mal group,the mice’s wei ght have different degree of losing compar ed to the period before the experiment.(2)The quantity of A group’s lavage is little, lavage’s color is clear and transparent; Synovial fluid of group B is muc h, the color isyellow and not pervious to light; The quantity of A C and D group’slavage is more than group A, color is yellow and pe rvious to light.(3)The results of White blood cells count: B group was obviously higher than that of A、 C、 D (p<0.01); Group C and gr oup D count results are similar, the comparison differ ence of t hese two group’s datawas not s tatistically significant (p>0.05).(4)TNF and IL-12of gr oup Bwas significantly higher than that of group A、 C、 D group (p<0.01).The count results of group C and gr oup is similar D,,the comparisondifference of thes e two groups was not statistically significant(p>0.05).Conclusion:“Tong Jing Huo li Tang” can reduce the white blood cells、 TNF、 IL-12of the back air bag synovitis. The mechanismin of it to cure joints synovial is mainly by reducing the WBC seepage,infla mmatory fa ctors (TNF, IL-12) content,and reduce the infla mmatory reaction. Its anti-infla mmatory effects are basically the same with “HuaMo Yan Ke Li”.
