| Chimonanthus grammatus M. C. Liu is a Chinese large evergreen shrub. It is theJiangxi endemic species, which is profitable to be exploited.In this study, we adopted ultrasonic-assisted extraction in room temperature withorganic solvents and organic solvent extraction, at last, we got ethanol extract,petroluem ether extract, ethyl acetate extract and n-Buoh extract.We adopted improved Ellman method to test the antiacetylcholinesterase activityof petroluem ether extract, ethyl acetate extract and n-Buoh extract of C. grammatusleaves, with the result that the three parts all have the inhibitory activity, but thebiggest one is ethyl acetate extract.Then we used the MTT method to test the inhibitory activity of overall ethanolextract, petroluem ether extract, ethyl acetate extract and n-Buoh extract of C.grammatus leaves to lung cancer cell lines A549,with the result that the overallextract owns certain inhibitory activity to the lung cancer cell lines A549,while theconcentration of the overall extract is between500ug/ml and1000ug/ml,it got goodinhibitory effect.In general,it is not obvious that the extract of C. grammatus leavescan strongly inhibit the proliferation of lung cancer cell.To explain the material basis of C. grammatus leaves,the research adoptedGC-MS method to test the chemical composition of petroluem ether extract with theresult that33comounds were identied. Then used column chromatography methods,reverse silica gel column and sephadex gel column to separate and purify the ethylacetate extract of C. grammatus leaves, we got7compounds in all.In addition to the physicochemical properties and pop characteristics, thechemical structures of4compounds have been identified. They are β–sitosterol(compound1), quercetin (compound3), kaempferol (compound4) and isofraxidin(compound6), respectively.The results of separation and purification provide basic data of certain chemicalto explain the composition of ethyl acetate extract. |
