| [Objective]Isolate and culture human chorionic cancer stem-like cells from human choriocarcinoma JEG-3cell lines with serum-free suspension culture, identify their biological properties in vivo and study the chemotherapeutic sensitivity.[Methods](1) Isolate and culture human chorionic cancer stem-like cells from human choriocarcinoma JEG-3cell lines with serum-free suspension culture;(2) Observe the tumor-initiating capacity in nude mice between JEG-3cancer stem-like cells and JEG-3cell lines in vivo experiment;(3) Observe the attacking ability by Transwell invasion in vitro experimental between the two groups of cells;(4) Detect the expression of OCT-4, NANOG by Real-time PCR between two groups of cells;(5) observe the chemotherapeutic sensitivity by MTT under different concentration of etoposide, fluorouracil and methotrexate between the two groups.[Results] (1)Human choriocarcinoma JEG-3cell lines can survive, proliferate and suspended cells with self-renewal capacity in the serum-free medium, and have the possibility of continuous extending.(2) JEG-3cancer stem-like cells have high tumorigenicity;(3) In the Transwell little chamber testing, JEG-3cancer stem-like cells have stronger invasive ability than JEG-3cell lines in vitro (P=0.006, P<0.05);(4) OCT-4, NANOG mRNA were higher in JEG-3cancer stem cells than in JEG-3cell line.(5) JEG-3cancer stem cells have higher survival rate and drug resistantance than JEG-3cell line; the chemotherapeutic sensitivity is statistical significance from low to high concentrations (P<0.05).[Conclusion]1. Human chorionic carcinoma JEG-3cancer stem-like cells have high tumorigenicity and high invasive ability than JEG-3cell line;2. Human chorionic carcinoma JEG-3cancer stem-like cells have high drug resistance, and OCT-4, NANOG mRNA were higher in JEG-3cancer stem cells than in JEG-3cell line, provide the theory basis for targeted treatment of choriocarcinoma for clinical,... |
