The Effects Of Xiao Ji Powder To Estrogen, Progestogen And Their Receptors Expression On Uterine Myoma Model Rat | | Posted on:2014-02-17 | Degree:Master | Type:Thesis | | Country:China | Candidate:H F Jing | Full Text:PDF | | GTID:2284330434458150 | Subject:TCM gynecology | | Abstract/Summary: | PDF Full Text Request | | Objectives:To observe the change of gross morphology of uterus of model rats by Xiaoji gra-nule and the influence on estrogen and progestin in serum and estrogen receptor (ER) and pro-gestin receptor (PR) in uterus tissue and discuss the mechanism of action ofXiaoji granule for prevention and cure of myoma of uterus by copying rat models with myoma of uterus and giving different doses of Xiaoji granule in contrast to intervention by Guizhifuling Capsule.Methods:72female rats were randomly divided into blank group, model group, positive control group, high-, medium-and low-dose Xiaoji granule groups. Except blank group, models of myoma of uterus were prepared with the method of synergistic induction by estrogen and pro-gestin in other groups. Pharmacological intervention was given since the ninth week. Serum E2, P, FSH and LH were measured after16weeks. The histopathological morphology of uterus was observed and uterus and ovary coefficients were measured and calculated. Meanwhile, ER and PR expressions of uterus tissue were observed with immunohistochemical method. The result was analyzed with statistical method.Results:Serum E2level of rats after modeling increases significantly and P<0.01, the difference has significant statistical meaning. Progesterone level decreases, but the difference doesn’t have statistical significance. Compared to model group, E2level of rats subject to positive pharmaco-logical intervention and intervention of different doses ofXiaoji granule is lower, but the differ-ence doesn’t have statistical significance. Compared to model group, the progesterone level of rats in groups subject to pharmacological intervention doesn’t change much. Estrogen and pro-gestin levels of rats in positive group and Xiaoji granule groups with different doses do not have difference with statistical significance. Through pairwise comparison of Xiaoji granule groups with different doses, estrogen and progestin levels of rats do not have significant difference. Modeling doesn’t have great influence on serum LH and FSH levels of normal rats. LH level of rats subject to positive pharmacological intervention and those inXiaoji granule groups with dif-ferent doses increases compared to model rats and the difference has statistical significance. LH level of rats in positive group and medium-doseXiaoji granule group increases significantly. FSH level doesn’t change significantly. LH and FSH levels of rats in positive group andXiaoji granule groups with different doses do not have significant difference; through pairwise comparison of-Xiaoji granule groups with different doses, the difference of serum LH and FSH levels of rats doesn’t have statistical significance. ER expression in uterus tissue of rat models with myoma of uterus increases compared to normal rats. P<0.05and the difference has statistical significance. PR expression increases significantly. P<0.01and the difference has significant statistical mean- ing. In positive group, ER expression decreases and PR expression increases, but the difference doesn’t have statistical significance. In low-doseXiaoji granule group, ER and PR expressions of model rats increase. PR expression increases significantly compared to model group and the dif-ference has statistical significance (P<0.05). In medium-dose Xiaoji granule group, ER level in uterus tissue of rats is inhibited. Compared to model group, it has statistical significance (P<0.05),but it doesn’t have great influence on PR expression. In high-dose Xiaoji granule group, ER expression increases compared to model group. The difference between them have statistical significance (P<0.05).
