Kai-Kou-Jian (rhizomes of Tupistra chinensis BAK.) is a reputed folk medicine herb for its potent to reduce carbuncles and to ameliorate pharyngitis. Primary studies showed that steroidal saponins, alkaloids, especial some cardiac glycosides are potentially active components of Tupistra chinensis. However, few of these compounds have been reported to be applied in treatment for immunological or inflammatory associated diseases. This study aimed to confirm the therapeutic effects of extraction on immunological liver injury and explore their possible mechanisms, mainly by examining its effect on PC1-DTH induced contact sensitivity, Con A and carbon tetrachloride-induced liver injury.In the first chapter of this study, we first extracted and examined the chemical composition. Tupistra chinensis were extracted by70%ethanol, and we got the extraction, which access rate is60.37%. And then, we examined the in vivo effect of PC1-DTH induced contact sensitivity, found that the extraction significantly suppressed the ear swelling. The pathological observations also showed that inflammatory infiltration induced by picryl chloride was improved by the extraction.In the second part of this research, we first examined the in vivo effect of the extraction on Con A-induced liver injury. The results showed that extraciton significantly inhibited the elevation of ALT, AST and LDH level in serum. The pathological observations showed that several pathological symptoms induced by Con A were improved by the extraction. Considering cytokines in the process of this model, we then examined their levels and found that the extraction markedly down regulated TNF-a and IFN-y in serum and liver tissue. We next tested the signal pathways, including NF-κB、STAT1and ERK changed, which indicating that extraction have a special way in treatments for immunological and inflammatory by control different proteins. We then examined carbon tetrachloride-induced liver model, found that extraction didn’t inhibit the elevation of ALT and AST level in serum. We confirmed that the extraction’s action in vivo was not the health protection for liver, but the effect to T lymphocyte.In the last Chapter, we demonstrated that Tupistra chinensis have effects on T lymphocyte in vitro. As a result, we found that the extraction had no toxicity and without affecting their activation to Con A stimulation, but it suppressed cytokines expression of activated T lymphocyte in vitro, and promoted apoptosis of Con A activated T lymphocyte in vitro.In summary, our research work all above is that immunological liver injury induced by Con A was improved by the extraction, which concerned that Tupistra chinensis improved the injury in immunological or inflammatory through suppressed the expression and induced apoptosis of T lymphocyte, also it was a pharmacological evidence for us to explore and untilize Tupistra chinensis.
