Expression Of Histon Deacetylase3in Placenta Syncytiotrophoblast With Intrahepatie Cholestasis Of Pregnancyand Its Significance

Objective To evaluate the expression and location of the histonedeacetylase3(HDAC3) in human placenta syncytiotrophoblast withintrahepatie cholestasis of pregnancy (ICP) and its significance.Methods From March2012to November2013, Thirty women withICP who delivered in the First Affiliated Hospital of Chongqing MedicalUniversity were chosen as the study objects.Thirty women wereclassifiedinto mild group (n=15), severe group (n=15) and elective term cesareansections group without previous onset of labor and perinatal complications(control group，n=15)．ELISA method was employed to examine the serumTNF-α. The expression and localization of HDAC3protein was detectedby Immunohistochemical. Use Western blot analysis to quantify HDAC3protein expression level．Examine serum total bile acid (TBA)、alanineaminotransferase (ALT) and aspartate aminotransferase (AST) byautomatic biochemical analysis instrument.Results （1）The TNF-α,TBA,ALT and AST levels in serum of the severe and mild groups were (52.35±3.16)ng/L,(69.95±21.47)ng/L,(312.2±34.7)U/L,(237.7±31.6)U/L and (39.08±2.27)ng/L,(11.31±0.76)ng/L,(101.7±19.2) U/L,(71.0±12.9) U/L, respectively, compared withmild group,serum TNF-α,TBA,ALT and AST levels wear higher in severegroup (P<0.05); they were both significantly higher than those in controlgroup (32.40±2.13ng/L,2.70±0.21ng/L,21.9±2.8U/L,17.3±1.3U/L,P<0.05)． And serum TBA,ALT and AST levels in mild group wearsignificantly higher than those who in control group (P<0.05).（2）In themild and severe ICP group,HDAC3was located in the placentasyncytiotrophoblastic cells, mostly in the cell endonuclear and stained lighttan; in control group, HDAC3was also located in placentasyncytiotrophoblastic cell stained dark tan and mainly in the cellendonuclear.（3）The HDAC3protein level in placenta syncytiotrophoblastof the severe groups and mild groups was0.077±0.006and0.076±0.007,respectively, compared with mild group, which showed no significantdifference(P＞0.05); they were both significantly lower than that in controlgroup (0.232±0.018, P<0.05)．（4）The quantitative expression of HDAC3in placenta syncytiotrophoblast was negatively correlated with serum TNF-α,TBA,ALT and AST levels,Spearman correlation coefficients were-0.340,-0.688,-0.533,-0.577.Conclusions （1） Down regulation of HDAC3expression of ICP inplacenta syncytiotrophoblast cells.（2） The quantitative expression of HDAC3in placenta syncytiotrophoblast was negatively correlated withserum TNF-α,TBA,ALT and AST levels（.3）The immune injury mediatedwith down regulation of HDAC3in ICP placenta syncytiotrophoblast maybe associated with liver function damage.
