| Aims:The model of vascular injured was established by balloon the rabbit carotidartery,to observe effects of TSG on TNF-αã€IL-6and IL-10protein expression inblood and vascular tissue injury site in rats after vascular balloon injury.Methods:The study of90male Wistar rats were obtained from the Experimental AnimalCenter of University of South China, weighing about300-400g,and randomly dividedinto the Sham-operated group, the injury group(The left common carotid arteryballoon injury)and the treatment group(The left common balloon injury+TSGgavage)with30in each, and the Sham-operated group only isolated carotid arterysurgery; surgery and treatment groups to build the carotid artery balloon injury model.Respectively take heart blood2ml after postoperative1d,4d,7d,14d and21d, eachtime point6rats, the blood samples were centrifuged to obtain supernatants, thendetecte TNF-α, IL-6and IL-10protein expression in serum-line by ELISA; The leftCarotid tissues of the three groups are harvested7ã€14and21days after taking heartblood,tissues are used to HE staining,observed the intimal and the VSMCproliferation and the ratio of intimal and medial by optical microscope;and thespecimens of7days for real-time PCR and western blot to detecte TNF-α, IL-6andIL-10protein expression.Results:1.The results of HE staining (To observe the ratio of intimal hyperplasia and intimal medial area by light microscope):Sham-operated rats carotid artery intima wassmooth, single layer and elastic wavy membrane,membrane was mainly smoothmuscle cell and the outer membrane was connective tissue.Compared with thesham-operated group,among modeling each time point,vascular intimal wasendarterectomy damage when surgery and treatment groups was1d and4d, butneointimal had not yet formated,IA/Ma was0;7d and beyond neointimal formationvisibled, and gradually thickening,cell proliferation was significantly, texturearranged disorder, extracellular matrix increased, wall thickened,which ultimately ledto vascular stenosis,suggesting vascular intimal hyperplasia after balloon injury;Whilethe treatment groups with vascular intimal hyperplasia,wall thickening and luminalstenosis was significantly reduced.HE staining results suggest that vascular intimalhyperplasia was concerned with endothelial injury and Stilbene glycosides cansignificantly reduce the vascular intimal hyperplasia and stenosis afer balloon injuryin rats.2. The results of ELISA:eath time point after surgery showed that the proteinexpression of TNF-a and IL-6in surgery and treatment groups were significantlyheighten compared with the Sham-operated group,while treatment lower than thesurgery group (P<0.05). Moreover protein expression of7d indicators of TNF-α, IL-6are highest.The protein expression of IL-10in surgery and treatment groups weresignificantly heighten compared with the Sham-operated group,further the proteinexpression of IL-10in treatment group was the most significantenhancement,Moreover protein expression of7d indicators of IL-10are highest.3. The results of real-time PCR:The mRNA expression of TNF-a and IL-6insurgery and treatment groups were significantly heighten compared with theSham-operated group,but treatment lower than the surgery group (P<0.05). ThemRNA expression of IL-10in surgery and treatment groups were significantlyheighten compared with the Sham-operated group,moreover treatment higher than thesurgery group (P<0.05).4.The results of Western Blot:The protein expression of TNF-a and IL-6in surgery and treatment groups were significantly increased compared with theSham-operated group,while treatment obviously lower than the surgery group(P<0.05). The protein expression of IL-10in surgery and treatment groups weresignificantly heighten compared with the Sham-operated group,further treatmenthigher than the surgery group (P<0.05).Conclusion:1. The vascular intimal hyperplasia was concerned with endothelial injury andStilbene glycosides can significantly reduce the vascular intimal hyperplasia andstenosis in balloon injury in rats.2. Stilbene glycosides can regulate inflammatory response by inhibiting theprotein expression of TNF-a and IL-6,as well as up-regulating the protein expressionof IL-10in plasma and vascular tissue after balloon injury in rats. |
