Objective:By measuring the CYP11B2-344C/T polymorphism and the relevant inflammationfactors, NF-кB and MCP-1, discuss the susceptible genotype and allele, as well as therelationship between the aldosterone synthase gene polymorphism-344C/T gene andthe inflammation factors concentration.Methods:During2012to2013,80cases of primary hypertension patients were chosen fromthe Department of Cardiovascular medicine, the Second Clinical College of Nan HuaUniversity, their blood pressure was between1to2grade.And the other80cases ofpeople from the Examination Center of the Second Clinical College of Nan HuaUniversity. All the people were between20to65years old and they were chosenrandomly.All the people were divided into four groups,40cases for eachgroup,essential hypertension with obesity group (group A) and essential hypertensiongroup (group B), they were chosen from the Department of Cardiovascular medicine,obesity group (group C) and healthy control group (group D),were chosen from theExamination Center. For these primary hypertension patients,we take the criteria"Guidelines for the Prevention and Treatment of Hypertension in China2010",andobesity diagnostic criteria use "Guidelines for Prevention and Control of Overweightand Obesity in Chinese Adult". The concentration of Plasma NF-кB and MCP-1wereboth measured by (ELISA). The CYP11B2-344C/T gene polymorphisms weredetected using Restriction Fragment Length Polymorphism (RFLP),PolymeraseChain Reaction(PCR) and agarose gel electrophoresis (AGE) methods. SPSS18.0software was applied for the data analysis. Results:1. The comparative analysis of the clinical dataThere were no significant difference on age, gender, fasting blood glucose (FBG)of the Aã€Bã€Cã€D four groups (P>0.05).The systolic blood pressure(SBP), diastolic blood pressure (DBP) of Group Aand B, as respectively compared with Group C and D,had significantdifference(P<0.05). And the body mass index(BMI), waist hip ratio (WHR), totalcholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C) andhight density lipoprotein cholesterol (HDL-C)of A Group and obesity group,asrespectively compared with B group and D group, had significant difference(P<0.05).2. The comparative analysis of CYP11B2-344C/T genotype frequencydistributionThe rate distribution of TT, TC and CC genotypes for each group: Group A were80%ã€12.5%ã€7.5%respectivelyï¼›Group B were70%ã€20%ã€10%respectivelyï¼›Group C were72.5%ã€17.5%ã€10%respectivelyï¼›Group D47.5%ã€32.5%ã€20%respectively. The genotype frequency among the four groups showed significantdifference (P<0.05). Compared with Group D,the rate of TT genotype of Group A, Band C were much higher, while the rate of TT+TC genotype were lower than GroupD. Had significant differences (x2value separately were9.14,4.18,5.21, P<0.05), andin Group A,the difference was showed to us the most significant.3. The comparative analysis of CYP11B2-344C/T allele frequencydistributionThe frequency of T and C alleles for each group: Group A were86.25%ã€13.75%respectively; B group were80%ã€20%respectively; Group C were81.25%ã€18.75%respectively; Group D were63.75%ã€36.25%respectively. The allele distributionamong the four groups showed significant difference (P<0.05)(x2value were13.23,P=0.004). Group A, B and C were multi-compared with the number of T and C allelesshowed significant difference (x2value separately were10.8,5.23,6.14, P<0.05), and Group A was was the most significant.4.The comparative analysis of plasma NF-кB and MCP-1concentrationof each groupGroup A, B and C compared with Group D respectively,the plasma NF-кB andMCP-1concentration had significant difference (P<0.05); Group A compared withGroup B and C respectively,the plasma NF-кB and MCP-1concentration hadsignificant difference (P<0.05); Group B compared with Group D,the plasma NF-кBand MCP-1concentration had no significant difference (P>0.05).5. The comparative analysis of MCP-1concentration in TT, TC and CCgenotypes of each groupGroup A, B and C compared with Group D respectively the MCP-1showedsignificant difference (P<0.05); Each group the concentration of MCP-1in TT werehigher than TC,CC genotypes significantly, except the MCP-1concentration in TTgenotype and TC Genotype. MCP-1concentration in TC genotype and CC genotypehad no significant difference (P>0.05).6. The comparative analysis of NF-кB concentration in TT, TC and CCgenotypes of each groupGroup A, B and C compared with Group D respectively, the concentration ofplasma NF-кB showed significant difference (P<0.05);. Each group the concentrationof NF-кB in TT were higher than TC, CC genotypes significantly. MCP-1concentration in TC genotype and CC genotype had no significant difference(P>0.05).But,in group D TC genotype was higher than CC genotype.Conclusions:1.The CYP11B2-344C/T polymorphism could be related with the pathogenesisof essential hypertension patients with obesity, and TT genotype may be susceptiblegenotype.2.The serum concentration of MCP-1and NF-кB in essential hypertension patients with obesity were higher,it suggested the existence of a certain extent TTgenotype of inflammation. |
