ObjectiveTo investigate the protective effects of carnosine on high glucose inducedapoptosis and its potential mechanism.MethodsThe SH-SY5Y cells were cultured by medium (DMEM) which containedwith high glucose50mmol·L-1and carnosine0.6、3or15mmol·L-1for48hours.The changes of cell morphology were observed by inverted phase contrast, cellviability was detected by MTT, changes of nucleus were assayed by Hoechst33258, apoptosis rate was measured by flow cytometry using Annexin-V andpropidumiodide (PI), levels of ROS in cells were detected by fluorescent probe,contents of8-OHDG in culture medium were determined by enzyme-linkedimmunosorbent assay.ResultsCompared with normal control group (low glucose DMEM), the highglucose group’s cell viability was decreased (P<0.01); karyopyknosis, split andapoptosis were found by Hoechst33258dyeing. The apoptosis rate and thecontent of8-OHDG were increased; Compared with high glucose group, the cellviability of carnosine (0.6,3or15mmol L-1) protect groups were obviouslyincreased (P<0.01); karyopyknosis and split form of cells significantlyimproved(P<0.01); apoptosis rate and the level of ROS were reduced (P<0.01); the contents of8-OHDG in culture medium were reduced(P<0.01). Comparedwith normal control group, the cell viability, change of nucleus and apoptosisrate were no significant change in carnosine, levels of ROS and contents of8-OHdG were respectively reduced(P<0.05). There were no significantdifferences between the normal group and the mannitol group.ConclusionsThe results show that carnosine has protective effect on high glucoseinduced apoptosis, the mechanism may be related to its antioxidation.
