Tumor is one of the main diseases to destroy human’s health and take away ourlives. Recently, operation and chemotherapy are reported as the main means ofmalignant tumor treatment. But clinical chemotherapy drugs not only damage the tumorcells but also impair the normal cells. Therefore, the treatment can’t be carried outsuccessfully.Traditional Chinese herbal medicine for treatment of tumor has a long history andabundant resources, which is enormous library of natural products. Natural productsplay an important role in the discovery of new drugs and lead compounds. BecauseChinese herbal medicine has the characteristics of diversity of chemical structures andbiological activities, it has become a crucial approach to screen out the efficient, low-toxic and cheap drugs which are used to develop anti-tumor drugs, and have becomethe research focus of attention in the new century.By investigating all the related papers in the world. We learn that the traditionalChinese herbal KZLA has extensive pharmacological activities, like antitumor,antioxidant, anti-HIV, anti-diabetes, antibacterial, etc. Among them, the anti-tumoreffect is valued.This research choose traditional Chinese herbal KZLA as research object. Guidedby the antitumor activity, choosing two kinds of human esophageal cancer cells (Eca-109and EC-9706) and through cell culture experiments of antitumor activity screening,we find that the chloroform extraction and ethyl acetate extraction have stronger anti-tumor activity. What’s more, we have screened out three hybrid ingredients and fouringredients respectively in chloroform extraction and ethyl acetate extraction.By using various chemical and chromatographic methods, separation andpurification work are performed smoothly. We have already gotten ten compounds fromethyl acetate extraction. They are5-hydroxy-1,4-naphthoquinone (1), β-sitosterol (2),5,7,3,4’-tetrahydroxy-flavone (3),3,4,5-trihydroy benzoic acid (4),3,5,7,3’,4’-penta hydroxy-flavone-3-O--L-rhamnoside (5),5,7,4’-trihydroxy-flavanone (6),3,5,7,3’,4’-pentahydroxyflavanone (7),3,5,7,3’,4’-pentahydroxy-flavone (8),4-hydroxy-3-methoxy-benzoic acid (9),3,5,7,3’,4’,5’-hexa hydroxy-flavone-3-O--L-rhamnoside (10).The other constituents, on account of low amount, still need to begathered, and their structures need to be determined. Due to the limited time, sevencomponents are separated from chloroform extraction, and chloroform extraction needs to be subdivided.This study is guided by the antitumor activity. It has screened out two activeextractions from traditional Chinese herbal KZLA, chloroform extraction and ethylacetate extraction. And then this research screened out three stronger active hybridingredients and four stronger active hybrid ingredients respectively from chloroformextraction and ethyl acetate extraction. In addition, this research is the first researchwhich uses human esophageal cancer cells as research object to study the anti-tumoractivities of traditional Chinese herbal KZLA. It provides references for further study ofanti-tumor activities research of traditional Chinese herbal KZLA.
