It is widely recognized by modern medicine that free radicals possess strongoxidability and thus could attack biomacromolecules, causing damages to tissues andcells and resulting in cancer, inflammation, cardiopathy, cardiovascular diseases anddiabetes, etc. Epidemiological studies have shown that intake fruit can reduce the riskof cancer and chronic disease. This paper takes polyphenols, flavonoids, anthocyaninsof persimmon, black plum and other five kinds of fruits as research object, detectingthe content of three kinds of active ingredients in seven kinds of fruits. We usechemical simulation and cell culture system to analyze the activity of antioxidantactivity in vitro, and anti-tumor proliferation of the extraction of the fruits. Thenexplore mechanisms of inhibiting tumor through flow cytometry and ELISAtechniques.Detect biological active ingredient content of seven fruit extracts, seven fruitextracts contain rich polyphenols, flavonoids and anthocyanin. The polyphenolcontent of persimmon is the highest,3.416mg gallic acid equivalent/g fresh weight.The flavonoids content of persimmon is the highest, of9.532rutin equivalent mg/fresh weight g. The anthocyanin content of black plum is the highest,1.569mgknapweed-3-glucoside equivalent/g fresh weight. The antioxidant capacities weresystematically analyzed through chemical simulation. By the analysis of totalreducing power, scavenging of free radicals(·OH, ABTS·+, DPPH·), persimmon wasfound to possess the lowest IC50value capacity, in the order:1.582mg/mL,0.845mg/mL,0.0450mg/mL. Persimmon was found to strongest antioxidant, followed bybayberry, black plum, raspberry, mangosteen, kiwi fruit and apricot. Additionally, itsantioxidant capacity had a strong correlation with its total polyphenol content(P<0.01).Furthermore, black plum was found to possess the lowest IC50value, in the order:747μg/mL,384μg/mL. Black plum was found the strongest ant-tumor capacity,followed by bayberry, raspberry, mangosteen, kiwi fruit, apricot and persimmon.Additionally, its anti-tumor capacity had a strong correlation with its total polyphenolcontent, total anthocyanin content (except for persimmon).Observe cell morphology of colon cancer Caco-2. Caco-2was found that cellmembrane edge was not neat, chromatin is rough, cell density decreased after treating by black plum extract. DNA Ladder experiment shows the characteristic of apoptosiscells gradient stripe. Results showed that the extraction of black plum couldefficiently induce Caco-2cells. More specifically, the extraction of black plum couldblock Caco-2at the stage of G2/M or G0/G1. At last apoptosis related proteins CytC,caspase-9, caspase3can be detected, so we can speculate apoptosis mediated bymitochondrial pathway. This indicated that the extraction of black plum could induceapoptosis in different paths to achieve the goal of proliferation of cancer cell.
