ObjectiveTesting the expression of Lats family in normal tubal epithelium, normal ovarian epithelial cells, ovarian mucinous cystadenoma, ovarian borderline mucinous cystadenoma, ovarian mucinous carcinoma and ovarian clear cell cancer and analysis their clinical significances. Exploring the origin of ovarian mucinous tumors and clear cell cancer by testing the expression of secretory cell marker PAX8 and mesothelial cell marker Calretinin in normal tubal epithelium, normal ovarian epithelial cells, ovarian mucinous cystadenoma, ovarian borderline mucinous cystadenoma, ovarian mucinous carcinoma, ovarian clear cell cancer, normal appendix, appendix cancer, normal colon, colon cancer, endometrium, endometrium cancer and endometriosis.MethodTesting the expression of Lats1, Lats2, PAX8 and Calretinin in 16 cases of normal oviduct epithelium, 14 cases of normal ovarian epithelium, 16 cases of ovariann mucinous cystadenoma, 14 cases of ovarian borderline mucinous cystadenoma, 18 cases of ovarian mucinous carcinoma and 9 cases of ovarian clear cell cancer by IHC. Counting the Immunohistochemical semi-quantitative score in every kind of ovarian tissues and percentage of positive cells of PAX8 and Calretinin in every kind of tissue in this study and analyze the data by using software SPSS 16.0 and compare the Immunohistochemical semi-quantitative score in each kind of ovarian tissue and the percentage of positive cells of different kind of tumor tissue with the normal oviduct epithelium with t-Test. Compare the expression of Lats in different clinical characteristics with Fisher’s Exact Test, Significant level α=0.05.Result1. Lats1, Lats2 were highly expression in normal oviduct epithelium and the percentages of positive cell of PAX8 in cell nucleus and cytomembrane of normal oviduct epithelium were(76.29±2.17)% as positive control while Calretinin were absolutely negative.2. Lats1, Lats2 were negative in normal ovarian epithelium and mucinous cystadenoma and moderately expression in ovarian borderline mucinous cystadenoma, but highly expression in mucinous cystadenocarcinoma. mucinous cystadenoma had no significance comparing with normal ovarian epithelium(P>0.05); ovarian borderline mucinous cystadenoma was higher than mucinous cystadenoma with P<0.05; mucinous cystadenocarcinoma was also higher than ovarian borderline mucinous cystadenoma with P<0.05.The exprisson state of Lats1 and Lats2 both had no significance in different clinical characteristics(P>0.05).3. The percentages of positive cell of PAX8 and Calretinin were 0% and(8.36±0.75)% in the 14 cases of normal ovarian epithelial tissues;the percentages of positive cell of PAX8 and Calretinin were(7.38±3.25)%, 0 in 16 cases of ovariann mucinous cystadenoma tissues, and the percentages of positive cell of PAX8 was lower the normal oviduct epithelium with statistically significance(P<0.05). The percentages of positive cell of PAX8 and Calretinin were(8.57±3.19)%, 0, in 14 cases of ovarian borderline mucinous cystadenoma tissues and the percentages of positive cell of PAX8 was lower compared with the normal oviduct epithelium with statistically significance(P<0.05). The percentages of positive cell of PAX8 and Calretinin were(85.00 ± 13.78)%, 0, in 18 cases of ovarian mucinous carcinoma tissues and the percentages of positive cell of PAX8 was higher compared with the normal oviduct epithelium with statistically significance(P<0.05). PAX8 and Calretinin were expressed in cell nucleus. Lats1, Lats2 were highly expressed in normal appendix and normal colon but barely expressed in appendix cancer and colon cancer, PAX8 was negative in normal appendix, appendix cancer, normal colon, and only 2 of the 19 cases were positive; Calretinin was all negative in these 4 tissues.4. Lats1, Lats2 were weakly expressed in ovarian clear cell cancer tissue.5. The percentages of positive cell of PAX8 and Calretinin were(86.44±10.82)%, 0 in 9 cases of ovarian clear cell cancer tissues and the expression of PAX8 had no statistically significance compared with normal oviduct epithelium(P>0.05) while Calretinin were negative. Lats1 and Lats2 were both highly expressed in endometrium, endometrium cancer and endometriosis; PAX8 was positive in endometrium, endometrium cancer and endometriosis; Calretinin was negative in all these 3 tissues.Conclusion1. Lats1 and Lats2 increased in normal ovarian epithelial cells, ovarian mucinous cystadenoma, ovarian borderline mucinous cystadenoma, ovarian mucinous carcinoma;2. The exprisson state of Lats1 and Lats2 both had no significance with different clinical characteristics;3. The origin of ovariann mucinous cystadenoma and ovarian borderline mucinous cystadenoma may still unclear;4. Ovarian mucinous carcinoma may partly originate from the the oviduct;5. Ovarian clear cell cancer probably mainly originate from the epithelium of fallopian tube.
