Study On Mechanism Of Compound Danshen Dripping Pills Combined With CBZ In The Treatment Of Kainic Acid Refractory Epileptic Rats | Posted on:2016-05-25 | Degree:Master | Type:Thesis | Country:China | Candidate:S S Han | Full Text:PDF | GTID:2284330461467161 | Subject:Pharmacy | Abstract/Summary: | PDF Full Text Request | Objective This papr attempts to study the possible antiepileptic mechanism of Compound danshen dripping pills by the change of GDNF, Bcl-2 and Bax expression in hippocampus detected by western blot based on the kainic acid refractory epilepsy rats model by the Compound danshen dripping pills, CBZ alone or integrated therapy, observing the kainic acid refractory epilepsy ratsbehavior, cognitive impairment and hippocampal neurons in order provide a new therapeutic method for combir traditional Chinese and western medicine treatment of refractory epdepsy.Methods1 The SD rats were divided randomly into saline control group, model group. Compound danshen dripping pills group, Compound danshen dripping pills combined with CBZ group. The saline control group was received saline. The other groups were given microinjection of kainic acid intracerebroventricularly. The seizure levels and ttie frequency of the spontaneous recurrent seizures were observed.2 7 rats were lected randomly from 45 d and 90 d groups perform Morris water maze(MWM)estl. The number of crossing the plat form and the timent in the target quadrant in probe trial were corded.3 3 rats lected randomly from 45 d and 90 d groups were under anaesthesia by chloral hydrate intraperitoneal injection. Nissl staining was performecHo observe the number of surving neurons in hippocampal CA3 area.4 Select 4 rats randomly from Id,45 d and 90 d groups. Western blot was used detect the expression of GDNFã€Bcl-2 and Bax.Results1 With the extention of treatment time, compared to KA group, the stage of seizures and SRS frequency were observably decreased in Compound danshen dripping pills combined with CBZ group (P<0.05). The study found that Compound danshen dripping pills could efficiently control epilepsy.2 With the extention of treatment time, Compound danshen dripping pills and Compound danshen dripping pills combined with CBZ could markedly increase the swimming frequency of crossing platform and percent time spent in the target quardrant, which illustrates that Compound danshen dripping pills and Compound danshen dripping pills combined with CBZ could improve the learning and memory markedly.3 With the extention of treatment time, compared to KA group, CBZ and Compound danshen dripping pills combined with CBZ groups has a larger number of surving neurons in hippocampal CA3 area (P<0.01), while the effect of CBZ group was less efficient than the combined group (P<0.01). Both CBZ and Compound danshen dripping pills combined with CBZ could evidently reverse the damage of hippocampal neuron in CA3 area induced by KA, while the later is more efficient.4 With the extention of treatment time, Compound danshen dripping pills could upregulate the expression of GDNF protein and ratio of Bcl-2 and Bax remarkably.ConclusionCompound danshen dripping pills combined with CBZ could more effectively control the epileptic seizures and improve the learning and memory, suggesting that the mechanism had some relationship with the improvement of hippocampal injury, the upregulated GDNF protein expression and the ratio of Bcl-2/Bax. | Keywords/Search Tags: | Compound danshen dripping pills, Carbamazepine, kainic acid, epilepsy model, Morris Water Maze, hippocampus, neurons, GDNF, Bcl-2, Bax | PDF Full Text Request | Related items |
