| Objective:We have found that miR-133b is under expression in gastric cancer and can regulate the expression of downstream gene SLC6A1 in previous studies. In this study, we explore SLC6A1 expression level in gastric cancer tissues and gastric cancer cells, then research the effect on the proliferation and apoptosis in gastric cancer cells when knock-down the expression of SLC6A1. Discuss the role of SLC6A1 in the process of gastic cancer. Simultaneously offers new reference for the future targeted therapy of gastric cancer and may serve as a molecular target in GC diagnosis and prognosis.Methods:Through the technique Real-time PCR detected the expression of mRNA in gastric cancer tissues and normal gastric mucosa epithelial cell GES-1 and gastric cancer cell lines AGS, SGC-7901, BGC-823, MGC-803, and though Western Blot detected the protein expression of SLC6A1 in above-mentioned cells. Down-regulate the SLC6A1 expression in above-mentioned cells using si-SLC6A1, detected the proliferation changes by CCK-8, and detected the apoptosis and cell cycle changes by FCM.Results:SLC6A1 mRNA expression lever is higher in gastric cancer tissues than in pericarcinomatous tissue. SLC6A1 mRNA and protein expression in gastric cancer cell and GES-1 from high to low is SGC-7901, BGC-823, MGC-803, AGS and GES-1. We knock-down the expression of SLC6A1 through RNA interference. After knock-down the expressions of SLC6A1, the proliferation of cells were suppressed, apoptosis were promoted. si-SLC6A1-transfected cells appeared to accumulated in S phase when compared to untransfected cells.Conclusions:1. The expression of SLC6A1 in gastric cancer tissues than para-carcinoma tissues, and it in gastric cancer cells was higher than that in GES-1, and SLC6A1 may be an oncogenesis gene.2. The highest expression of SLC6A1 in gastric cancer cells is in the middle differentiation cell SGC-7901, the least is in high differentiation cell AGS, SLC6A1 may take part in the process of cell differentiation.3. SLC6A1 can promote cell proliferation and inhibited of cell apoptosis in gastric cancer cells, and SLC6A1 also change the cell cycle of gastric cancer cells. |
