ObjectiveStudy the kidney deficiency postmenopausal osteoporosis’histone demethylase JMJD2 family expression and their impact to histone methylation (H3K9me3, H3K36me3) levels. Futher study their correlation with ERR a, To establishment the links between histone demethylase JMJD2 family and TCM ’kidney deficiency’postmenopausal osteoporosis. Providing a new method for the prevention and treatment of osteoporosis from both Traditional Chinese Medicine and Western Medicine.MethodsBetween October 2014 and December 2014, We select 20 postmenopausal women, whose age between 50-70 years old, take THA due to osteoarthritis of the hip, they all came from The First Affiliated Hospital of Guangzhou University of Traditional Chinese Medicine Orthopedics Center. After detect lumbar spine BMD,10 patients with kidney deficiency postmenopausal osteoporosis as our study group, and 10 without osteoporosis as control group. All the 20 patients were tested peripheral venous blood 25-OH VitD level, during THA, we collect the cancellous bone specimens from femoral head, then immunohisto-chemistry, Western blot assaying bone tissue ERR a, JMJD2A, JMJD2B, H3K9me3 and H36me3 expression. We use the SPSS 18.0 software package for the data statistical analysis. The results were considered signif ican at P<0.05. Student’s test was used to determine the significance of study parameters on a continuous scale among groups. The Pearson correlation analysis was used to study the correlation. All data were described as mean it standard deviation(x±SD).Results1. Both of the two group’s ERRα, JMJD2A and JMJD2B expression from weak positive to positive, for postmenopausal osteoporosis group, the expression level was lower than that non-osteoporosis group, there was a statistically significant difference between the two groups(P<0.05); In postmenopausal osteoporosis group, the H3K9me3, H3K36me3 expression levels higher than that non-osteoporosis group, there was a statistically significant difference between the two groups(P<0.05).2. The JMJD2A, JMJD2B expression is positively associated with ERRa,25-OH Vit D, both reduced expression in postmenopausal osteoporosis patients.Conelusion1. For postmenopausal women, The histone demethylase JMJD2A and JMJD2B expression level is differ, in postmenopausal osteoporosis patients’, the JMJD2A and JMJD2B expression was significantly reduced, with the corresponding that postmenopausal osteoporosis patients’histone methylation level (H3K9me3, H3K36me3) elevating, suggesting that histone methylation is associated with osteoporosis, while histone demethylase JMJD2A and JMJD2B reduce the occurrence of osteoporosis.2. The histone demethylase JMJD2A and JMJD2B have a positive corres-pondding with ERRa, Vit D, the specific transport mechanism remains to be elucidated.3. Histone demethylase JMJD2A and JMJD2B may serve as a’kidney deficiency’ postmenopausal osteoporosis’antagonistic enzyme.
