Objective:To study the effects of acupuncture combined with enriched rehabilitation training on the sensory and motor function and expression of nerve growth factor(NGF) and brain derived neurophic factor(BDNF) in rats after focal cerebral ischemia injury so as to explore the effect and mechanism of acupuncture, enriched rehabilitation training and comprehensive treatment in recovering ischemic brain damage.Method:Rat models of local cerebral ischemia were established by thread- occluding the middle cerebralartery(MCAO).The rats were randomly divided into shamoperated,model,acupuncture,enriched rehabilitation trainingandacupuncture combined enriched rehabilitation traininggroups. each group was divided into 3subgroups by 3 d、7 d and 14 d,The marks of neural function defect、sensory and motor ability were noted down separately. At3 d,7dand 14 d ofcerebral ischemia injury,the immunohistochemistry staining was applied to observe the expression of NGF and BDNF cells in rats. The expression of NGF and BDNF were analysed after cerebral ischemia among the above groups.Results:1.The sham operated group had no nerve function deficiency symptoms,neuronal function evaluation of rats intreatment groups and model group had no obvious difference before therapy(P>0.05). neuronal function evaluation of rats in treatmentgroups and model group had reduce trend aftersurgery, at 4h,3d ofcerebral ischemia injury,neuronal function evaluation of rats in treatment groups and model group had no obvious difference(P>0.05).at 7d,14 d ofcerebral ischemia injury,the evaluation of neuronal function in treatment groups lower than model group, the improvement of the acupuncture combined enriched rehabilitation training group was significant(P<0.01),the improvement of acupuncture group and enriched rehabilitation training group were remarkable(P<0.05).the marksof neuronal function evaluation of acupuncture combined enriched rehabilitation traininggrouplower than acupuncture group and enriched rehabilitation training group,the improvement of acupuncture combined enriched rehabilitation training group was remarkable(P<0.05);at 4h, 3d,7d and 14 d ofcerebral ischemia injury,neuronal function evaluation of acupuncture group andenriched rehabilitation training grouphad no obvious difference(P>0.05).2. The evaluation of rotating bar,ofrotating bar and net screen in treatment groups and model group had reduce trend after surgery,At 4h,3d ofcerebral ischemia injury,the evaluation of rotating bar, ofrotating bar and net screen in treatment groups and model group had no obvious difference(P>0.05).At 7d ofcerebral ischemia injury,the evaluation of rotating bar, ofrotating barand net screen in treatment groups lower than model group, compared with model group,the evaluation of rotating bar, ofrotating barand net screen in acupuncture combined enriched rehabilitation training group was significant(P<0.01),the marks of rotating bar, ofrotating barand net screenevaluation ofacupuncture group and enriched rehabilitation training group were remarkable(P<0.05);the evaluation of ofrotating bar of acupuncture combined enriched rehabilitation training group lower than cupuncture group and rehabilitation group,compared with acupuncture group and enriched rehabilitation training group, the marks of ofrotating barevaluation of acupuncture combined enriched rehabilitation training group was remarkable(P<0.05), at 14 d of cerebral ischemia injury,the evaluation of rotating bar,ofrotating bar and net screen in treatment groups lower than model group,compared with model group, the marks of rotating bar, ofrotating barand net screen evaluation of acupuncture combined with enriched rehabilitation training group was significant(P<0.01),the marksof ofrotating bar evaluation of acupuncture group was significant(P<0.01),the marks of rotating bar and net screen evaluation of acupuncture group was remarkable(P<0.05),the marks of rotating bar, ofrotatingbarand net screen evaluation of the enriched rehabilitation training group was remarkable(P<0.05),the evaluation of ofrotating bar, net screenin acupuncture combined enriched rehabilitation training groups lower than acupuncture group and enriched rehabilitation traininggroup,compared with compared with compared with acupuncture group and enriched rehabilitation training group,the marks of ofrotating bar evaluation of acupuncture combined enriched rehabilitation training group was significant(P<0.01),the marks of net screen evaluation of acupuncture combined enriched rehabilitation training group was remarkable(P<0.05),at 4h,3d,7d and 14 d ofcerebral ischemia injury,the marks rotating bar, ofrotating barand net screenevaluation of acupuncture group andenriched rehabilitation training grouphad noobvious difference(P>0.05).3. After cerebral ischemia injury, the expression of NGF and BDNF increased more obviously.at 3dcerebral ischemia injury,positive expression of NGF and BDNFof treatment groups and model group reached to a peak. at 7d ofcerebral ischemia injury,positive expression of NGF and BDNF of treatment groups and model group were downward trend at 14 d ofcerebral ischemia injury,positive expression of NGF and BDNF of treatment groups and model group continued to reduced(P<0.01).at 4h, 3d,7d and 14 d ofcerebral ischemia injury,compared with model group, the expressions of NGF and BDNF were increased and strengthen in ischemia cortical areas of three treatment groups(P<0.01).The three treatment groups compared with one another,which showed that the acupuncture combined rehabilitation group had more significant effect on promoting the expressing of NGF and BDNF(P<0.05).Conclusion1.The therapy of acupuncture combined rehabilitation promoted rehabilitation of neuronal function after focal ischemia,and it was better the therapy of simple enriched rehabilitation trainingand the acupuncture.2. The therapy of acupuncture combined rehabilitation increased thigh muscle strength, improved walking, balance and grasping ability of cerebral infarction in rats,and it was better the therapy of simple enriched rehabilitation trainingand the acupuncture.3. acupunctureandrehabilitation can increase the expression of BDNF and NGF protein, and acupuncture combind rehabilitation training can improve more significantlyto neurological recovery.acupuncture combind rehabilitation trainingto improve the function mechanism of focal cerebral ischemia injurymay be related to the promotion expression of NGF and BDNF in of cerebral ischemia injury.
