Changes Of Vaginal Microenvironment In 20 Women After Placing NuvaRing Contraceptive Ring

Background Vaginal contraceptive ring is a new-style contraceptive way used in the last decade.The most widely-used vaginal contraceptive ring in foreign country is NuvaRing at present-NuvaRing is the first vaginal contraceptive ring approved for use by U.S. Food and Drug Administration.For contraceptive effect as good as oral contraceptive, easy and convenient operation, high bioavailability,few side effects,it is deeply affected by foreign women.In China,we have started to study this ring,however,we have made little study of vaginal microenvironment in using vaginal contraceptive ring. Vaginal microenvironment is so significant that we could not ignore it. Once it is destroyed or out of balance, vagina inflammation could happen in women.Now there are 20 women recruited by Qi Lu Hospital.By colleting vaginal discharge and vaginal lavage of taking contraceptive ring before and after,we do a series of research in them.Objective To study the changes of vaginal microenvironment in 20 women before and after taking the vaginal ring 1,3,6 cycles in vagina.Method Among 20 women who had used vaginal contraceptive ring, Vaginitis five joint inspection and Microscopy were used to examine the vaginal discharge of different cycles;the concentration of IL-2 and IL-8 in vaginal lavage of all subjects were determined by enzyme linked immunosorbent assay (ELISA).Results The concentration of IL-2 and IL-8 before taking the ring is 7.68±1.44pg/mL、 277.07±56.01pg/mL.The concentration of IL-2 and IL-8 after placing the ring one cycle is 7.95±1.40 pg/mL、347.50±68.40pg/mL.Compared to the ring placed before,there was a significant difference in some large after taking the ring one cycle(P<0.05); the concentration of IL-2 and IL-8 after placing the ring 3 cycles is 7.66±1.41 pg/mL, 291.21±41.62 pg/mL; the concentration of IL-2 and IL-8 after placing the ring 6 cycles is 7.6711.33 pg/mL,287.94144.29 pg/mL. The concentration of IL-2 and IL-8 droped and retured to normal during this time(P>0.05). Through Vaginitis five joint inspection and Microscopy,the pH,quantification of leukocytes,lactobacilli,BV (bacterial vaginosis),VVC (vulvovaginal candidiasis),TV(trichomonal vaginitis)of vaginal discharge had no significantantly difference compared to the ring placed before.Conclusion There was some effects on the concentration of IL-2 and IL-8 after placing vaginal contraceptive ring initially,but they can return to normal along with the ring adopting to vagina and self-regulation of body; furthermore,vaginal ring had no obvious effects on vaginal acid circumstances and couldn’t lead to the growth of other bacteria.
