| Objectives : Tumor associated macrophages(TAMs) is one of the important factors in the body against tumor. At the same time, it has series functions of promoting the development of tumor. It includes supporting for the proliferation, infiltration and metastasis of tumor cells. So there is high correlation between TAMs and the tumor patients with poor prognosis.This study uses immunohistochemical Envision method to analyze the the level of TAMs and the expression of VEGF165, bcl-2 in 62 cases of breast carcinoma and 40 cases of adjacent normal tissue. After that, it combined with the clinical pathologic characteristics and follow-up data, and discusses the impact of TAMs,VEGF165 and bcl-2 in the process of development of breast carcinoma, and to study the correlation of TAMs,VEGF165 and bcl-2.Methods: This study uses simple random sampling method and collects 62 breast carcinoma samples which were surgically removed and have holonomic pathologically diagnosis with and complete follow-up data in Bethune international Peace Hospital from January 2010 to December 2010, at the same time all the patients did not receive any radiotherapy and chemotherapy before their surgery, meanwhile it collected 40 cases of adjacent normal tissue to breast cacinoma as comparison. Afterwards it uses immunohistochemical Envision method to detect the level of TAMs which is marked by CD68 and the expression of VEGF165 and bcl-2 in breast carcinoma and adjacent normal tissues. The study used SPSS16.0 software package for statistical analysis and processing. In breast carcinoma tissue and adjacent normal tissue, the count of TAMs and expression of VEGF165 and bcl-2 are measurement data. If each data was normal distribution, it should be shown insx ±. If it was skewness data, it should be shown in the median(interquartile range),(M(QR)). The count of TAMs,the expression of VEGF165 and bcl-2 and the relationship between the clinical pathologic data which use Student’s t test, wilcoxon rank sum test and inspection or Kruskal Wails inspection analysis. It analyze on Spearman Rank Correlation Coefficient about the count of TAMs, the expression of VEGF165 and bcl-2 in breast carcinoma tissue.Rusults:1 TAMs were marked with CD68. They have been expressed in breast carcinoma and in adjacent normal tissue with cytoplasm dyed buffy. They have been expressed in the stroma of breast carcinoma largely(M = 25.2). A small amount of distribution was in normal tissue adjacent to carcinoma(M = 0.93). VEGF165 and bcl-2 were dyed buffy in the cytoplasm. They have been expressed in tumor cells largely(M = 5.06, M = 2.56), A small amount of expression was in normal tissue adjacent to carcinoma(M = 0.40, M = 0.60). Compared to the adjacent normal tissue, TAMs, VEGF165 and bcl-2 were highly expressed in breast carcinoma tissue, with statistically significantly difference(P < 0.05).2 The count of TAMs in group age at 50 years and older was significantly higher than in group at less than age 50 years old(P = 0.048), there was no statistical significance of the expression of VEGF165 and bcl-2 between the the two age groups(P > 0.05).3 The count of TAMs and the expression of VEGF165 in C-erb B2 positive group was significantly higher than C-erb B2 negative group(P < 0.05), there was no statistical significance of the expression of bcl-2 between C-erb B2 positive group and C-erb B2 negative group in breast carcinoma(P = 0.864).4 The count of TAMs and the exression of VEGF165 in vascular tumors bolt group was significantly higher than no vascular tumors bolt group(P < 0.05), there was no statistical significance of the expression of bcl-2 between the two groups(P = 0.073).5 The count of TAMs and the expression of VEGF165 in gradeâ…¡and â…¢ groups were significantly higher than in gradeâ… group(P < 0.05), there was no statistical significance of the count of TAMs and the expression of VEGF165 between gradeâ…¡group and grade â…¢ group(P > 0.05). there was no statistical significance of the expression of bcl-2 among the three groups(P = 0.041).6 The count of TAMs and the expression of VEGF165 and bcl-2 in lymph node metastasis group was significantly higher than that without lymph node metastasis group(P < 0.05).7 There was no statistical significance of the count of TAMs and the expression of VEGF165 and bcl-2 between survival duration of less than 3 years patients group and survival duration of more than 3 years patients group(P > 0.05).8 In breast carcinoma tissues, the count of TAMs and the expression of VEGF165 exist positive correlation(P < 0.05), Spearman correlation coefficient r = 0.714;The expression of VEGF165 and bcl-2 exist positive correlation( P < 0.05), Spearman correlation coefficient r = 0.264; The count of TAMs and the expression of bcl-2 exist positive correlation(P < 0.05), Spearman correlation coefficient r = 0.281.Conclusion:1 The count of TAMs, the expression of VEGF165 and bcl-2 in breast carcinoma was significantly higher than in adjacent normal tissue.2 In breast carcinoma tissues, TAMs and VEGF165 can promote tumor lymph node metastasis,and promote the progression of tumor.In breast carcinoma tissues,bcl-2 may promote tumor lymph node metastsis.3 In breast carcinoma tissues, TAMs and VEGF165,the count of TAMs and the expression of bcl-2,the expression of VEGF165 and the expression of bcl-2 were all of positive correlation. TAMs through the secretion of VEGF on promoting tumor angiogenesis, at the same time to raise the the level of bcl-2 by VEGF, inhibiting the apoptosis of tumor cell and promote the progression of tumor. |
