| Objective: To investigate the expressions of human epidermalgrowth factor receptor-2(HER2),epidermal growth factor receptor(EGFR) and S100A14 in colorectal carcinoma, adenoma and normal mucosa tissue by immunohistochemistry. Further to explore the relationship between different proteins and colorectal carcinoma; Then, to analyze the association between HER2, EGFR, S100A14 proteins and clinical features, to discuss the significance of the proteins in the development of colorectal carcinoma. This study aimed to discuss the role of HER2, EGFR, S100A14 and the possible mechanism in the development of colorectal carcinoma,and to provide more reliable methods and indicators for clinical targeting therapy.Methods: 120 cases of colorectal carcinoma, 84 cases of adenoma and normal mucosa tissue were collected between 2012 and 2013 from Tangshan Gongren Hospital. Moreover, we studied the expressions of HER2, EGFR, S100A14 by immunohistochemistry. The analysis of results were dealed with related statistical methods.Results:1 The positive rate of HER2 protein was 70.00% in colorectal carcinoma tissue, 14.28% in adenoma tissue, 11.90% in marginal mucosa respectively. The positive rate of HER2 in colorectal carcinoma tissue was obviously higher than those two groups,and there were both significant differences X2=61.564,P=0.000; X2=67.120,P=0.000).There were no differences in colorectal carcinoma tissue and marginal mucosa(X2=0.209,P=0.647).2 The positive rate of HER2 was 57.57% and 85.18% in well and moderate differentiation, worse differentiation respectively, there were significant differences(X2=10.781, P=0.001); The positive rate of HER2 with and without lymphatic metastasis were 88.89% and 54.54% respectively, there were significant differences(X2=16.681, P=0.000). 81.35% and 59.01% in invasive depth with serosa and without of serosa respectively, there were significant differences(X2=7.127, P=0.008).60.31% and 80.7% in TNM stage â… + â…¡and â…¢+ â…£respectively, there were significant differences(X2=5.921, P=0.015). But the expression of HER2 was not related with ages, sex, and neoplasm location(P>0.05).3 The positive rate of EGFR was 65.83% in colorectal carcinoma tissue, 57.14% in adenoma tissue, 35.71% in the marginal mucosa tissue respectively. The positive rate of EGFR in colorectal carcinoma tissue was obviously higher than that in marginal mucosa tissue, and there were significant differences in this two groups(X2=18.015,P=0.000). The positive rate of EGFR in adenoma tissue was higher than that in marginal mucisa tissue,and there were also differences in the two groups(X2=7.754,P=0.005). There were no differences in colorectal carcinoma tissue and adenoma tissue(X2=1.588,P=0.208).4 The positive rate of EGFR was 51.56% and 82.14% in well and moderate differentiation, worse differentiation respectively, there were significant differences(X2=12.417, P=0.000); The positive expression of EGFR with and without lymphatic metastasis were 87.03% and 51.51% respectively, there were significant differences respectively(X2=17.083, P=0.000), 81.35% and 50.81% in invasive depth with serosa and without of serosa respectively, there were significant differences(X2=12.433, P=0.000).57.14% and 75.43% in TNM stage â… + â…¡and â…¢+ â…£respectively(X2=4.453, P=0.035). But the expression of EGFR was not associated with ages, sex, and neoplasm location(P>0.05).5 The positive rate of S100A14 protein was 66.67% in colorectal carcinoma tissue, 66.67% in adenoma tissue, 45.24% in the marginal mucosa tissue respectively. The positive rate of S100A14 in colorectal carcinoma tissue was obviously higher than that in marginal mucosa tissue, and there were significant differences in this two groups(X2=9.305,P=0.002). The positive rate of S100A14 in adenoma tissue was higher than that in marginal mucisa tissue,and there were also differences in the two groups(X2=7.825,P=0.005). There were no differences in colorectal carcinoma tissue and adenoma tissue(X2=0.000,P=1.000).6 The positive rate of S100A14 was 50% and 85.17% in well and moderate differentiation, worse differentiation respectively, there were significant differences(X2=17.143, P=0.000); The positive expression of S100A14 with lymphatic metastasis and without lymphatic metastasis were 81.48% and 54.54% respectively, there were significant differences respectively(X2=9.796, P=0.002), 79.66% and 54.09% in invasive depth with serosa and without of serosa respectively, there were significant differences(X2=8.819, P=0.003). 50.79% and 84.21% in TNM stage â… + â…¡and â…¢+ â…£respectively, there were significant differences(X2=15.038, P=0.000). But the expression of S100A14 didn’t have association with ages, sex, and neoplasm location(P>0.05).7 Positive correlation between HER2 and EGFR(r=0.28, P=0.002), HER2 and S100A14(r=0.309, P=0.001), EGFR and S100A14(r=0.564, P=0.000)were found in colorectal carcinoma. Conclusion:1 HER2, EGFR and S100A14 were obviously overexpressed than that in mucosa in colorectal carcinomas, indicating HER2, EGFR and S100A14 play roles in the different process of mucosa to adenocarcinoma.2 The levels of HER2, EGFR and S100A14 expression were closely associated with worse differentiation, LNM, inflated deeply and TNM â…¢+â…£,suggesting that they might be involved in the invasion and metastasis processes.3 HER2, EGFR and S100A14 protein levels were associated with each other, which they might interact synergistically to play their functions. |
