| Objiective: Acne Vulgaris is a kind of skin disease of unknowm etiology.Its pathogenesis is complex,oxidative stress may be one of the important in the pathogenesis.Oxidative stress is due to active substances in cells to produce too much,more than the removal of the antioxidant ability,leading to the oxidation of biological macromolecules of suger,lipid and protein.But the oxidation-antioxidant imbalance associated with the debelopments of acne vulgaris,has not been reported.Therefore,this study proposed by determination of serum in patients with acne vulgaris and normal control grou Pof oxidation and antioxidation of related indicators:Protein carbinyl(PC),malondialdehyde(MDA),nitricoxide(NO), glutathione(GSH) an d superoxide dismutase(SOD) expression levels,and investigate the pathological mechanism of oxidative stress in acne vulgaris and the role of disease development,provide the experimental basis for clinical treatment.Methods:1 The experimental groupCase group: Acne patients including 96 patients(50 females and 46 males) with an average age of 20.3 years,were all Han population. According to the Global Acne Grading System(GAG), according to the disease activity is divided into the following three groups:(1) mild acne(2) moderate acne(3) severe acne.The normal control group:Control grou Pconsisted of 90 healthy subjects(48 females and 42 males); mean age was 21.2 years old,were all Han population.No statistical dfference between the two groups of objects in the age,gender and race.2 Specimen collectionFor the two groups were fasting venous blood collected elbow, procoagulant 5ml, low-speed centrifugation serum was collected and stored in aliquots of serum-20 ℃ refrigerator.3 Determination of oxidation indexThe determination of Protein carbonyl(PC) ã€malondialdehyde(MDA) and nitric oxide(NO) This three indicators are used ELISA double antibody sandwich method.4 Determination of antioxidant indexDetermination of glutathione(GSH) and Superoxide dismutase(SOD) This two indicators are used ELISA double antibody sandwich method.Results: 1 The oxidation level of acne vulgaris patients and normal control group 1.1 Protein carbonyl(PC) contentThe average protein carbonyl detection of acne vulgaris patients and normal control serum protein(PC) levels were 2.61±0.13 nmol/L and 2.08±0.09 nmol/L, acne vulgaris patients serum PC content increased significantly(P<0.05). The average PC content in the mild, moderate and severe acne vulgaris patients were 2.15±0.04 nmol/L, 2.78±0.09 nmol/L and 3.23±0.12 nmol/L, compared with the normal control group, moderate and severe acne patients PC content was significantly increased(P<0.05), and mild acne patients without significant change(P>0.05) 1.2 The content of malondialdehyde(MDA) contentThe average malondialdehyde detection of acne vulgaris patients and normal controls(MDA) content in serum were 5.11±0.28 nmol/ml and 4.58±0.19 nmol/ml, MDA content in serum of patients with acne vulgaris were significantly increased(P<0.05). The average MDA content in the mild, moderate and severe acne vulgaris patients were 4.81±0.19 nmol/ml, 5.26±0.21 nmol/ml and 5.48±0.16 nmol/ml, compared with the normal control group, mild, moderate and severe acne patients the average MDA content significantly increased(P<0.05). 1.3 nitric oxide(NO) contentThe average nitric oxide detection of acne vulgaris patients and normal controls(NO) content in serum were 46.53±2.17μmol/L and 28.79±1.09 μmol/L, NO content in serum of patients with acne vulgaris were significantly increased(P<0.05). The average NO content in the mild, moderate and severe acne vulgaris patients were 40.23±1.21μmol/L, 48.36±1.59μmol/L and 55.82±1.49 μmol/L, compared with the normal control group, mild, moderate and severe acne patients the average NO content significantly increased(P<0.05). 2 acne vulgaris patients and normal control grou Pthe antioxidant level 2.1 glutathione(GSH) contentThe average glutathione detection of acne vulgaris patients and normal controls(GSH) content in serum were 37.54±1.63 ng/L and 44.02±1.47 ng/L, acne vulgaris patients serum GSH content was significantly reduced(P<0.05). The average GSH content in the mild, moderate, severe acne vulgaris patients were 41.55±0.76 ng/L, 35.82±0.63 ng/L and 32.63±1.08 ng/L, compared with the normal control group, moderate and severe acne patients with GSH was significantly lower(P<0.05), and the average GSH content in patients with mild acne did not significantly decrease(P>0.05). 2.2 supero xide dismutase(SOD) content The average superoxide dismutase detection of acne vulgaris patients and normal controls(SOD) content in serum were 34.51±0.25 U/m L and 37.50±0.19 U/m L, the average SOD content in serum of patients with acne vulgaris were significantly reduced(P<0.05). The average SOD content in the mild, moderate, severe acne vulgaris patients were 35.08±0.18 U/m L, 34.18±0.12 U/m L and 33.90±0.16 U/m L. Compared with normal control group, mild, moderate, severe acne patients the average SOD content decreased significantly(P<0.05)Conclusion:1 The existence of oxidant antioxidant imbalance in the serum of 1 patients with acne vulgaris. Oxidation level increased, the antioxidant level significantly decreased.2 moderate and severe oxidation antioxidation in serum in patients with acne vulgaris imbalances than mild acne vulgaris patients. |
