Based on three-dimensional fluorescence data combined with second order correction algorithm, new methods for the determination of chelerythrine, a medicinal component of Chinese herbal medicine Chelidonii Herba(Baiqucai), and for the determination of nitidine chloride, a medicinal component of Zanthoxyli Radix(Liangmianzhen), were established. Problems involved in the methods, such as the correction of the fluorescence inner filter effect, the spectral properties of the materials being tested, and the algorithms, were investigated. The HPLC method was used to verify the accuracy of the new method.Three parts were included in this thesis:Fluorescence inner filter effects within small molecule systems were studied. Using L-tryptophan as a fluorophore, coumarin or 6-hydroxyflavanone as light absorption materials, the primary and secondary inner filter effects produced by the two materials, respectively, were investigated. It was indicated that the decline of logarithm of fluorescence intensity lg F was in direct proportion to the concentration of the light absorption materials. When the absorbance A of the light absorption materials was less than 0.05, fluorescence inner filter effect disappeared.2. Fluorescence spectra and absorption spectra of chelerythrine were studied. Influences of experimental conditions on the fluorescence of chelerythrine were tested. Fluorescence quantum yield of chelerythrine alkaline solution was measured to be 0.12. The dissociation constants of chelerythrine was determined to be p Ka = 8.36, by a p H-absorption method. The alternating trilinear decomposition(ATLD) algorithm was used to deal with the three dimensional fluorescence data matrix(Ex-Em-c), and to calculate the content of chelerythrine in a Baiqucai sample, the results of standard addition method and standard curve method were 0.0175% and 0.0172%, respectively. The result obtained by a HPLC method was 0.0179%.3. According to the effect of p H on the fluorescence spectra of nitidine chloride and coexisting components ethoxychelerythrine and toddalolatone in Chinese herbal medicine Liangmianzhen, a new method for the determination of nitidine chloride in Liangmianzhen crude drug was established, baded on three dimentional fluorescence data with second order correction algorithm. In this method, fluorescence excitation spectra of Liangmianzhen extraction and nitidine chloride standard solutions of different concentrations and different p Hs were measured, the fluorescence data were constituted to form a p H-Ex-c matrix, and then the alternating penalty trilinear decomposition(APTLD) algorithm was used to deal with V the matrix, and to calculate the content of nitidine chloride in Liangmianzhen sample. The new method was used to determine the contents of nitidine chloride in reference crude drug and in crude drug obtained from drug store. The reliability of the new method has beenverified by a HPLC method.
