ObjectiveTo study the origin pre-treating and processing integration techniques of Sophora flavescens Ait., and to explore its applied feasibility.MethodsTo analyze the situation of the traditional process and the origin pre-treating method, the quality evaluation system of S. flavescens Ait.was optimized and improved.To ensure the quality evaluation system could reflect the inner quality and it is easy for practice, First, Optimization method of Thin Layer Chromatography(TLC) and High performance liquid chromatography(HPLC) was used to qualitatively identify and quantitatively determine matrine, sophoridine and oxymatrine, respectively. Second, the traditional appearance shape and water-soluble extract were incorporated into the quality evaluation system. On this basis, using the orthogonal design method to optimize the tradition process of S. flavescens Ait., and single factor experiments to establish the cutting process parameters of fresh S. flavescens Ait. Furthermore, the industrialized production technique of the cutting process of fresh S. flavescens Ait was explored by combining with production practice and optimized process in the laboratory.Results1. Optimized the method of TLC was used to qualitatively identify matrine, sophoridine and oxymatrine: the testing solution and standard was dotted on silica gel G plate with chloroform-methanol(5: 0.2, v/v) which saturated by a mmonia vapor as the mobile phase., put it in After launching and took out air dries, spots were visualized by spraying with Wagner’s reagent followed by heating. The testing solution showed the same orange spot with standards, including matrine, sophoridine and oxymatrine.2. HPLC method simultaneous determination of matrine, chromatographic conditions sophoridine and oxymatrine: the assay was performed on Thermo-NH2(250 mm * 4.6mm, 5 μm) column, acetonitrile-ethanol- phosphoric acid(80: 10: 10) as the mobile phase, flow rate at 0.8m L· min-1, the wavelength at 210 nm, the temperature was 40℃,the injection volume was 10 μL.3. Eatablished the traditional processing optimum parameters: Took the dry herbs,removed residual root head, sized washed. soaked 30 min(the water temperature of about 20℃). After moistening to penetrate, cutting into 3 mm thick slices, 80℃ dried.4. The optimized cutting process of fresh S. flavescens Ait: the fresh herbs, put the drum-type washing machine to remove the sand, sun-dried or low temperature(50-60℃) dried to moisture of 38%-50%,cut 3-4 mm thick slices, 60-70℃ dried.5. Explored the industrialized production technique of the cutting process of fresh S.flavescens Ait: the fresh Sophora herbs, put the drum-type washing machine to remove the sand quickly, After sun-dried 7-12 days or low temperature(50-60℃) dried to moisture of 38%-50%, cut 3-4 mm thick slices with high speed reciprocating machine,60-70℃ dried.Conclusion1. The evaluation quality system is accurate and reliable for controlling of the oral and herb of S. flavescens Ait.2. Optimize the tradition process of S. flavescens Ait. would guide and standardize the effects of herb’s production enterprises. But it is difficult to guarantee uniform softening for many factors affected in the soften link. Such as,the size of S. flavescens Ait.,texture, air temperature and humidity, the temperature of soaking water, water consumption and so on.3. The cutting process of fresh S. flavescens Ait.for basis on the single factor experiment, which is easy to operate, and the quality is stable.4. Industrial production technology in the study of origin on the basis of the formation process suitable for large-scale, industrial production, improve quality Pieces, has certain social and economic value, will promote the process of standardization ofChinese Herbal Medicine.
