Effect Of Bushen Antai Granules On Decidualangiogenesis Of Recurrent Spontaneous Abortion Mice

Objective: Study on effect of Bushen Antai dissolved medicine on The blood vessels,cell morphology,vascular endothelial cell ultrastructure of recurrent abortion of mice decidual tissue. From the aspects Angiogenesis and so on,we explore the further mechanism of Bushen Antai Granules on pregnancy and fetal prote-ction. and we can use this as the basis for subsequent animal experiments. ortion, laying the theoretical foundation for further animal experiments in the future.Methods: We use Clark model which has a classic use of implicit, iterative and paternal specific characteristics of DBA / 2XCBA recurrent abortion mice model. The female CBA / J mice were with male DBA / 2 and BALB / c mice were mated by 2:1 were established RSA model CBA / J × DBA / 2 model with a normal pregnancy CBA ×BALB / c, male and female 2:1 housed separately vaginal plug detection of pregnancy were counted as 0 days. Animals were divided into six groups of 10 normal pregnancy group, model control group were given distilled water; comparison group to group administered progesterone capsules(0.4mg / kg.d); traditional Chinese medicine(Bushenantai granules) High, middle and low-dose groups, respectively 3.9g / kg.d,11.7g / kg.d, 35.1g / kg.d, distilled water orally, each group 15 days. After the last administration 24 h, the mice were sacrificed, cut deciduas remove.,vascular and cell morphological changes of decidual tissues were observed under light microscope,observation on the ultrastructure of vascular endothelial cells.The expression of decidua MVD was detected by immunohistochemistry(microvascular used as a marker CD34).Results: ① We use light microscope to observe, the number of vessels in the model group decreased cytoplasmic decidua severe edema, some nuclear disappeared karyopyknosis; progesterone group vascular rich decidua cytoplasmic swelling,vacuolation, few nuclei disappear; high medicine dose group and western medicine group microscopic structural similarity; traditional Chinese medicine, the low-dose group optical microscope relative reduction in the number of vessels, edema decidua cytoplasmic vacuolization.②Under electron microscope, model group incomplete vascular wall endothelial cells coated structure is incomplete, the nucleus was inside spindle, rough endoplasmic reticulum, ribosomes reduced mitochondria have been destroyed; progesterone group intact vascular wall, vascular structural integrity of endothelial cells, the nuclear membrane integrity, cell gap widened slightly, seen in the cytoplasm abundant rough endoplasmic reticulum and ribosomes, mitochondria swelling and degeneration of the structure; high-dose group and western medicine group microscopic structural similarity;traditional Chinese medicine, the low-dose group of vascular wall integrity, some endothelial cell swelling, nuclear membrane integrity, the cytoplasm of rough endoplasmic reticulum, ribosomes slightly reduced, partly mitochondrial swelling,vacuolization.③ Compared with normal pregnancy significantly reduced the expression of MVD decidua control mice, and was statistically significant(P <0.05). Compared with the control group, MVD expression of Chinese dose group and progesterone group were significantly increased, and a statistically significant(P <0.05 or P <0.01). Compared with the progesterone group, the expression of high dose group were no significant differences between the MVD(P> 0.05). Compared with the high dose group, the lower MVD expression levels of dose group of Chinese middle-dose medicine and Chinese low-dose group, The difference was statistically significant(P <0.05).Conclusion: Recurrent miscarriage mice exist deciduas that the number of blood vessels reducing, decidual cells and endothelial cell structure be severely damaged,organelles incomplete, MVD expression insufficient. Bushen Antai dissolved medicine can repair of the damage cellular structure of decidual tissue of recurrentabortion in mice,improving angiogenesis and cell organelle integrity angiogenesis,promoting embryonic development,it also plays a role in prevention of recurrent spontaneous abortion.
