120 Kunming male mice with 18 to 22g were randomly divided into six groups, one of which used for distilled water control group, one of which used for Pseudostellaria Polysaccharides control group,one of which used for cyclophosphamide (CY) group,and the rest for treatments,pseudostellaria polysaccharides control group and three treatmental groups were given at the dose of 400 mg/kg body weight, 100mg/kg body weight,200 mg/kg body weight and 400 mg/kg body weight to each mouse respectively by gastrogavage for 19 successive days,CY was give to each mouse both CY group and three treatment groups by intraperitoneal injection with 100 mg/kg body weight in the 20th day. Mice were sacrificed after 24 h.The spleen lymphocyte proliferation was detected by MTT Carbon clearance assay the phagocytic activity of macrophages, ELISA was used to detect serum cytokine (interleukin-2, interleukin-6, interferon-γ)levels, RIA assay the content of secretory immunoglobulin A (SIgA) and intestinal cytokines (interleukin-2,interleukin-6) in duodenum and ileum, Observation on the morphology of small intestine tissue and count the number of immune cells by microscope, focusing on the impact of immune regulation and immune protection protective effect of the intestinal mucosa Pseudostellariae polysaccharides on immune injury mice which was induced by CY. The results were as follows:(1) Compared with the CY model group, the high dose group of pseudostellaria polysaccharide was increased in thymus and spleen index index (P<0.05), Heterophylla polysaccharide group spleen index increased significantly (P<0.01), Heterophylla polysaccharide group thymus index increased significantly(P<0.05), between the dose groups, the thymus index and spleen index was increased with the dose increased.(2) Compared with CY model group, the index a of phagocytic was increased in the high dose group of Pseudostellaria polysaccharide (P<0.05), the index a of phagocytic was increased significantly in the control group of Pseudostellaria polysaccharide(P<0.01), and with the increased of the dose, the trend was increased, The OD values of blank hole, ConA hole and LPS hole has increased in the high dose group of heterophylla polysaccharide (P<0.05) and the OD values of ConA hole has increased in the middle dose group of heterophylla polysaccharide (P<0.05).The OD values of blank hole and LPS hole has increased significantly(P<0.05),the OD values of ConA hole has increased significantly in heterophylla polysaccharide control group(P<0.01).(3) Compared with CY model group, radix pseudostellariae polysaccharides of high, middle dose group mice serum IL-2 concentration increased (P<0.05), and radix pseudostellariae polysaccharide in high dose group mice serum IL-6, IFN-γ concentration increased significantly (P<0.05). In the control group of heterophylla polysaccharides, the content of IL-2 and IL-6 has increased in the serum of mice(P <0.05), IFN-γ content increased significantly (P<0.01).(4) Compared with CY model group, Each dose group of heterophylla polysaccharide and heterophylla polysaccharide group the intestinal villus length increased, the V/C ratio, Intestinal intraepithelial lymphocytes and the quantity of goblet cells were increased(5) SIgA contents duodenum were significantly increased (P<0.05) in middle and high treatment groups compared to the CY model group, SIgA contents ileum were significantly increased (P<0.01) in high treatment groups compared to the CY model group. IL-2 contents duodenum and ileum were significantly increased(P <0.05), IL-6 contents duodenum were significantly increased (P<0.01), IL-6 contents ileum were significantly increased (P< 0.05) in high treatment groups compared to the CY model group. In the control group of Heterophylla polysaccharide, SIgA and IL-2 and IL-6 were increased.This experiment through the establishment of immune injury model, found the intragastric administration of Pseudostellaria polysaccharide has certain protection on mice immune injury, The results showed that each dose group of Pseudostellaria heterophylla polysaccharides increased immune organ index; Increased macrophage phagocytosis index; Acts in synergy with ConA, LPS, antagonistic the decreased of spleen lymphocytes by CY; Increased serum levels of cytokine IL-2, IL-6, IFN-y content; there are some affect in improve the intestinal villus length and the ratio of V/C, And have some affect in improve the number of the muscular layer thickness and the number of intraepithelial lymphocyte and goblet cells increased; With the increase of dose effect is more significant; In intestinal mucosal immunization, Pseudostellaria heterophylla polysaccharides can improve the intestinal mucosal immunity antagonize IL-2, IL-6, SIgA content has decreased which was resulted by CY, And With increasing doses, the effect is more obvious.the result showed that Pseudostellaria heterophylla polysaccharides play a protective effect on intestinal mucosa. And the results of heterophylla polysaccharide group compared with the results of control group,indicated that pseudostellaria polysaccharides has a certain effect to improve the above indexes,which explained that heterophylla polysaccharide has a good effected in improved the body’s immunity.
