Objective: to establish three kinds of rats models of pulmonary artery hypertension, abdominal aortocaval fistula shunting(A-VF), morocrotaline(MCT)and MCT+A-VF, and set up control group. Exploring the role and their correlation of survivin, caspase-3 in pulmonary artery hypertension of rats.Methods: 40 SD rats were randomly divided into four groups, each group of 10, A- VF, MCT, MCT + A- VF and the control group. The m PAP and RVHI were detected; The change of small and medium pulmonary arteries was observed by HE. The expression of survivin and caspase-3 was detected in three kinds of PAH modles by the method of immunohistochemical.· Results:· 1. The m PAP and RVHI of three kinds of PAH modles are higher in the control group(P < 0.01), the MCT+ A-VF group increased more than the MCT group and A- VF group(P < 0.01), while the MCT group is higher than A- VF group(P < 0.01).· 2. The small and medium sized pulmonary arteries walls were normal in the control group. Arterial wall intima was smooth. There is no inflammatory cell in the lung tissue surrounding the arterial walls. The small and medium pulmonary artery walls thickened in the three kinds of models. the disordered arrangement of endothelial cells and smooth muscle cells, cell proliferation and luminal stenosis, they occured in the model groups. There were a lot inflammatory cells in the lung tissue surrounding the arterial walls of model groups. The MCT + A- VF group changes the most.· 3. The survivin expressed in the inner and outer membrane of arteris and the surrounding in the three kinds of PAH modle groups, did not appear in the control group. The number of possitive cells and tinting strength in the MCT + A-VF group were more than in the MCT group and A-VF group(P < 0.01),the MCT group was higher than A- VF group(P < 0.05).· 4. The caspase-3 expressed in the inner, middle and outer membrane of arteris and the surrounding in four groups, the control group was positive for high strength than the model groups(P < 0.05), the A-VF group was higher than the MCT group and MCT + A-VF group(P < 0.05), the MCT group is higher than the MCT + A- VF group(P < 0.05).· 5. The correlation was negative between the expression of survivin and caspase-3( r=-0.755,P<0.01).Conclusions:1. The increase of pulmonary artery pressure and remodeling of pulmonary artery are in the three kinds of model,which the MCT + A-VF group is heaviest.2. Survivin and caspase-3 are involved in the formation of pulmonary hypertension. The expression of survivin is up-regularion with the increase of pulmonary hypertension, and the expression of caspase-3 is down-regularion.The correlation was negative between the expression of survivin and caspase-3.
