| Objective Effect of resveratrol on unilateral ureteral obstruction rats renal interstitial fibrosis and delay the intervention study of residual renal function damage.Methods 24 clean grade SD rats were randomly divided into sham operation group(A group) and operation group(B,C),with 8 rats in each group,operation group rats by unilateral ureteral ligation method to establish animal model of renal interstitial fibrosis, divided into model control group(group B),resveratrol alcohol treatment group(group C). Are to be ordinary feeding, sham operation group and the model control group:Aã€B to 10ml/Kg normal saline,resveratrol treatment group: 25mg/kg.d of resveratrol(dissolved in saline to 10ml/Kg) intragastric administration, all rats were killed 14 days after operation,were examined after fourteenth days of serum urea nitrogen(BUN),creatinine(Cr),and rats were observed 14 days after UUO renal fibrosis:1) the dyeing method using HE and Masson to observe the changes of renal tissue and basal fibrosis in renal tubular epithelial cells;2) expression was assayed by immunohistochemistry method of TGF- β1,Col I,Smad2/3,Smad7 prote.Result 1) fourteenth days after operation,serum urea nitrogen,creatinine:model group than in sham operation group rats blood urea nitrogen,creatinine increased significantly(P<0.05),add resveratrol, rats blood urea nitrogen,creatinine decreased significantly(P<0.05).2) Model group compared with the sham operation group rats renal interstitial injury index was significantly increased(P<0.01);after resveratrol treatment, compared with model group, the renal interstitial injury index was significantly lower(P>0.05) than those in the sham operation group,the renal interstitial injury index was increased(P<0.01). 3) the pathological section Masson staining:the model group significantly increased collagen content(P< 0.01),and after resveratrol treatment,collagen deposition significantly reduced(P<0.01). 4) immunohistochemistry rat TGF-β1,collagen I,Smad2/3, Smad7 display: Sham operation group of renal interstitial no expression of type I collagen, model shows that a large number of type I collagen deposition,after resveratrol treatment, compared with model group,the expression of renal interstitial collagen type I significantly reduced(P<0.05);sham operation group rats kidney tissue TGF- beta1 weakly positive expression,model group TGF- beta1 expression was significantly higher than those in the sham operation group(P<0.01), resveratrol treatment group TGF- beta 1 expression than those in the model group decreased significantly(P<0.05);sham operation group rats Smad2/3 in renal tissue weak positive expression,model group showed strong positive expression,whereas resveratrol treatment group(P< 0.01) the reduced expression of Smad7 in renal tissue;sham operation group was obviously enhanced compared with the model group,while resveratrol treatment group than those in the model group increased expression of Smad7(P<0.01).Conclusion Resveratrol can be influenced by TGF- β /Smads pathway in Smad2/3,Smad7, TGF- β1 expression,intervention of renal interstitial fibrosis,Thus delaying renal failure. |
