Objective:By observing the influence of YiShenJiangZhuo Prescription to the signal path of chronic renal failure rats TGF-β/Smads to investigate the curative effect mechanism of YiShenJlangZhuo Prescription on chronic renal failure.Method:Divide the 60 clean 7 week-old male SD rats into groups including blank group,sham operation group, CRF model group, Nlaoduqing granule group, YiShenJiangZhuo Prescription group, and 12 rats for per group. Adopt the 5/6 nephrectomy to build the CRF rats model, and after 4 weeks.begin to give different medications and last for 8 weeks and then observing the index of serum Scr, BUN, β2-MG and urinary 02-MG and 24h urine protein quantitative on the different groups. We use immunohistochemical method to detect the expression of Smad3, Smad7, TGF-β1 protein in the renal tissue, and bserving the residual kidney pathological morphological changes by the light microscope.Result:1.In CRF rats model group BUN and Scr increased significantly (P<0.05), the expression of TGF-β1, Smad3 elevated and the expression of Smad7 reduced obviously (P<0.05), renal interstitial fibrosis is visible under the light microscope.2.Both YiShenJiangZhuo Prescription and Nlaoduqing granule are able to reduce the Scr, BUN, β2-MG and 24h urine protein quantitative(P<0.05)of the CRF rats, and reduce the expression of renal tissue TGF-β1, Smad3, and improve the expression of Smad7 (P<0.05) It shows that the renal interstitial fibrosis is relieved and the effect of YiShenJiangZhuo Prescription is better than Nlaoduqing granule.Conclusion:1. YiShenJiangZhuo Prescription is able to protect the function of renal tubular and glomerular, improve the renal function and delay the renal tissue fibrosis.2.The anti fibrosis mechanism may be related to inhibit the expression of TGF-β1, Smad3 and improve the expression of Smad7. It works By influencing the TGF-β/Smads signal pathway transduction to achieve the goal of delayed renal interstitial fibrosis and the CRF progression.
