Lumbar disc herniation, which threated human’ health life seriously was the most common form of spinal diseases. With changing of lifestyle and habits,the incidence rate of Lumbar disc herniation increased year by year, and having a trend of more and more young. Off tipe of prolapsed and free was a special type of of disc herniation free type, which not only can cause nerve root symptoms, but also can compress cauda equina easily. We could’t cure this tipe peitient according non-surgical treatment,but we could get a good result by surgical treatment, it can promptly remove the prominent organizations to release,the pressure on spinal nerve roots. however,in linic,there were a lot of patient, belonging this tipe, refused Surgical treatment.Some patients who adhere to conservative treatment result in symptoms relieved or even cured amond the same tipe patients of Lumbar disc herniation. In spite of it there were some documents thowing that disc prolapse prone can reduce by self-absorption, but there was no more evidence allow us easily decide to take a conservative treatment program.In order to avoid serious consequences, such as cauda equina compression. Therefore, to find enough evidences to support conservative treatment is necessary for these patients.Objective: To investigate the correlation between MMP-3 levels in blood and local implanted tissue,and the weight’changing of implanted tissue by testing content of MMP-3 of blood and local implanted tissue, and the weight of implanted tissue in SD rats erector spinae,a modeling of simulating prolapsed disc herniation, to explore the applying of MMP-3 in the conservative treatment of patients with disc herniation. Methods : fifty SD rats(only 3 months of age) were Randomly divided into normal control group, negative control group(including 45 days group and 60 days group), experimental group(including 45 days group and 60 days group), 10 in normal control group,and 20 in each group of the laters. The MMP- 3 content in ocal implanted tissue and the blood wre detected with enzyme-linked immunosorbent assay method after 45、60 days of experiment. Record the weight of implanted tissue before implanting and different time after oporation.Results:(1) The expression of MMP-3 was significantly higher in the experimental group and negative control group compared with that of normal control group(P<0.05);(2) 60 days group had a higher concentration of MMP-3 compared with 45 days group in both local organizations and blood, the difference was statistically significant, the former implanted organizations’ s self-absorption was faster than the latter..(P<0.05)(3)At at different time after surgery of experimental group, MMP-3’schange of local implanted organizations was highly positive correlation with the one in blood,and the correlation coefficients were 0.835 and 0.895, both P <0. 05.Conclusions: The content of MMP-3 in implanted tissue continued to rise after implantation, in the process of implanted tissue’s resorption would be faster with the increase of MMP-3 level;content of MMP – 3 in implanted tissue is highly correlated with the level of that in the blood; patients with disc herniation can come to understand the content of prominent local tissue of MMP-3 and its change by detecting the blood’ MMP-3 to provid a reference for treatment option.
