| ObjectiveTo investigate the effect of cordycepin on the mice with cyclophosphamide-induced myelosuppression by observing the change rules of signs, numbers of peripheral blood cells and femoral bone marrow nucleated cells and the expression levels of CENP-B in white blood cells.Methods30 mice of which weight were(25±2.5)g were randomly and averagely divided into three groups, that is control group, myelosuppression group and intervened by cordycepin group. Mice in control group were intraperitoneally injected of normal saline(NS) for 12 d with the dosage of 5m L·kg-1; Mice in myelosuppression group were injected of NS for 5d after injection of cyclophosphamide for 7d with the dosage of 50mg·kg-1; Mice in intervened by cordycepin group were injected of cordycepin for 5d with the dosage of 25mg·kg-1 after injection of cyclophosphamide for 7d. The methods of peripheral blood cell count, HE staining, western blotting and RT-PCR were applied to analyse the number of peripheral blood cells,the quantity and pathogeny structure of femoral bone marrow nucleated cells and the expression levels of CENP-B in white blood cells of the mice in all groups.ResultsThe changes of signs showed that mice in control group had a prompt move, a normal diet and a intense reaction to the sudden sound; The mice in myelosuppression group with the darker hair than those of the mice of control group had a slow reaction to the sudden sound, a dulled appetite and a slow growth in weight, meanwhile they showed the symptoms of gathering, huddling up, shivering and hematuresis. And after injected cordycepin, the mice in intervened by cordycepin group had a active action, a normal diet and a lustrous hair. Besides, the symptom of hematuresis disappeared. Compared with the indicators of the mice in control group, the numbers of leukocytes, red blood cells, platelets and nucleated cells, the content and the expression level of CENP-B of the mice in myelosuppression group were significantly reduced(P<0.05); And Compared with the mice in myelosuppression group, all the indicators except for the number of red blood cells have significantly recovered.Pathological structure of bone marrow nucleated cells showed the significantly reduction of the number of BMNCs and the presence of many bubbles in myelosuppression group which were effectively improved after injecting cordycepin. The results of Western blotting and RT-PCR showed that the relative content and the expression level of CENP-B in myelosuppression mice were lower than normal mice, whereas those of the mice in intervened by cordycepin group were picking up.ConclusionCordycepin has a certain therapeutic function to the mice with cyclophosphamide-induced myelosuppression mice. And this study will provide a data support to further explore the action mechanism of cordycepin. |
