| Objective To observe the expression and significance of the Platelet ultrastructure of acute cerebral infarction, platelet-related parameters MPV, PLT, PDW and platelet GMP-140, platelet-activating factor (PAF).Methods30patients with infarction of the cerebral median artery whose onset duration were between6-24hours, and30healthy adults cases as a control group, to observe the Platelet ultrastructure of the patients with transmission electron microscopy; Automatic blood cell analysis instrument measure MPV, PLT and PDW of the patients;Double-antibody sandwiching ELISA method measure the GMP-140, and platelet-activating factor (PAF) concentration of Plasma,then measure NIHSS scores of the30patients at onset and14days later. Aftet the antiplatelet treatment;its influence on relevant indicators of platelet was observed, and correlation analysis was made for NIHSS and platelet-related parametersResults The ultrastructural changes of platelets was evident with more pseudopodium,aggregation and fusion, more disruption of platelet membrance, fewer alpha granules and also mitochondria,the remains of which demonstrated swell and vacuolization. The results showed that MPV, expressions of GMP-140ã€PAF of all the patients increased and PLT was reduced notably with intervention; After intervening, MPV, GMP-140ã€PAF and NIHSS were reduced significantly, bur there is no obvious change for PLT;Compared with the PLT before treatment,there is no difference in statistics; Correlation analysis shows, MPV and PLT do not have statistics dependence on NIHSS,while GMP-140and PAF are correlated with NIHSS positively.Conclusion These observations showed that the presence of increased platelet function are associated with ultrastructural changes of platelets including more disruption of platelet membrance, significantly fewer alpha granules and also mitochondria, swelling of the remains of mitochondria. Platelet activation is one of important pathology changes of acute cerebral infarction, and it increased the danger of thrombogenesis. GMP-140and PAF have been increased in acute cerebral infarction,and decreased significantly after antiplatelet therapy,it can be indicated that its activation may be involved in the pathogenesis of cerebral infarction. |
