Initial Study On Extraction Of Lagenaria Siceraria Polysaccharide And Its Immunocompetence

Polysaccharide is composed by the polymerization of many a monosaccharide molecules and is widelyexists in the cell wall of plants,animals and microorganisms.It has been found that the polysaccharidecompounds has hundreds of chemical structural modls,with the development of biotechnology andchemical purification technology more and more chemical structure and pharmacological action were beingfound and it has been applied in the medical care ect.At present,it have been sure that the pharmacologicalacctions of different the polysaccharide are different and they show a phenomenons of various biologicalactivities,becuase of the differance of its structure,molecular weight and solubility.Based on the predecessorto the hoist in Uyghur medicine and medical application as foundation,select young (immature) Lagenariasiceraria as test material,firstly by using the orthogonal test method hoist the optimal extraction conditionsof polysaccharides,then extracted from each grade of polysaccharide and total polysaccharide,and studiedthe immune activity in vitro and vivo.The main results are as follows:1.The best extraction conditions of LSP was extraction temperature90℃,extraction time was120min,ratioof material to liquid extraction agent1:25,under the condition of extraction quality up to the LSP129.9mg/g, sugar content of41.64%.2.The LSP can accelerate the immune function of mice.Use of water decoction and alcohol extractionmethod by four different LSP in certain concentration range With different degrees of promotingproliferation of spleen lymphocytes in mice. Furthermore,In a certain range of concentration of LSP cansignificantly enhance the ConA induced lymphocyte proliferation.And more than certain concentrationrange,LSP could inhibit ConA induced lymphocyte proliferation.3.Studies of LSP on mouse peritoneal macrophage phagocytosis effect have shown that.LSP could enhancethe phagocytosis of macrophage.the80%high dose group and30%high dose group enhancement isobvious.4.Study on the effect on serum IL-2and TGF-β1mice concentration LSP show that LSP in certain doserange can significantly increase the content of IL-2in serum of mice,At the same time,reduce the serumTGF β1level of mice,Which play a role in the regulation of immune.5.Study on the effect of LSP on erythrocyte immune function shows that the LSP Can significantly improvethe rosette formation rate of normal mice red cell C3b receptor,increasing of normal mice erythrocyteimmune function.
