| Objective:In order to further clarify the effect of PNS Panax Notoginseng Saponins in the treatment of pulmonary fibrosis in the in vitro culture of human embryonic lung fibroblast cell line MRC-5,and to explore the mechanism o fPNS Panax Notoginseng Saponins intervention on MRC-5 cell proliferation,apoptosis,oxidative stress,cytokines and collagen and other aspects,in order to provide experimental basis for Chinese herbal medicine in the treatment of pulmonary fibrosis.Methods:According to the requirements of the project design,selection of human embryonic lung fibroblast MRC-5 as the research object,with PNS Panax Notoginseng Saponins for drug intervention,low molecular weight heparin drug control,the experiments were divided into three groups: control group(group A),low molecular weight heparin group(B group),PNS Panax Notoginseng Saponins group(C group).In vitro culture of human embryonic lung fibroblast MRC-5 cells,according to the groups were given corresponding intervention effect of CCK-8 method to detect different concentrations of PNS Panax Notoginseng Saponins on cell proliferation,according to the results of CCK8 to choose the optimal concentration of drugs,Annexin-V APC/7-AAD double staining method to detect cell apoptosis,expression of cultured cells was detected by ELISA in liquid TGF-beta 1,NF-ΚB CTGF,the content changes of MDA,SOD detection of cell biochemistry(cell protein),the changes in QPCR cell was detected by TGF-mRNA and NF-ΚB mRNA beta 1,CTGF mRNA,the content of Sirius red staining of collagen.Results and Conclusion:PNS Panax Notoginseng Saponins of certain concentration can inhibit human embryonic lung fibroblast MRC-5 cell proliferation,promote cell apoptosis,inhibit the secretion of collagen and CTGF,NF-kappa B,TGF-beta 1 and mRNA,decrease the content of MDA and increase the content of SOD.The study showed that total saponins of Chinese herbal medicine extract PNS Panax Notoginseng Saponins can affect the human embryonic lung fibroblast cells through a variety of ways,hinder the proliferation and transformation of the deposition of extracellular matrix fiber cells,has a certain anti pulmonary fibrosis. |
