| Lung cancer is one of malignancies that has the highest incidence and mortality which has caused huge losses to the society, and heavy grief to the family. During the recent decades, people have always focused on the study of lung disease. With the development of medical technique, the medical means are becoming more various, many lung cancer patients receive effective treatment. Owing to the late discovery, no scientific diagnosis, timely treatment, it is undeniable that for many patients with lung cancer miss the opportunity that can be cured. Therefore, it has become a critical issue for early detection, early diagnosis and early treatment to lung cancer.As known, the body is always going on metabolism, the metabolic pathways, the type and quantity of various subjects are in the state of dynamical balance. The variations of metabolites directly reflect the status of the physical body. Therefore, it has aroused considerable attention to study the pathogenesis and diagnosis of lung cancer based on the metabolites. Protein constructs the basis of life metabolic. As the basic unit of protein, the amino acids involve the synthesis of many enzymes, hormones, etc. So it is of great importance to study amino acid metabolism.Using high performance liquid chromatography (HPLC) and pre-column derivatization, Preliminary study on five amino acids in human serum and rat liver tissue has been made. The main contents are as follows:1. The feeding of nude mice and its preparation of tissue samples. The control group and the group with human lung cancer cells inoculated were feed under the condition of SPF. The tissue were selected when the tumor diameter was up to aboout5mm.2. The effective pretreatment of the samples of the human serum and rat liver tissue. In the morning, the blood of normal human, patients with lung cancer and lung cancer chemotherapied were collected. Then we adopt the centrifugal first, and then water bath coagulation method for collecting serum effectively, and the removing fibrinogen in ordor to prevent hemolysis. The rat liver tissue was made into homogenate through ultrasonic cell crusher, the conditions of ultrasonic pulverization are set as the power of200W, the working time of4s, intermittent time of10s, the ultrasonic frequency of40times.3. Accurate optimization of experimental parameters for simultaneous determination of amino acids in human serum and rat liver tissue samples through high performance liquid chromatography(HPLC). The optimized conditions include chromatography and derivatization, and the interference experiments of other amino acids. The optimized conditions are the acetonitrile-phosphate buffered saline solution of pH7.0, column temperature of37℃, detection wavelength of466nm. Meanwhile, the method provided is of high accuracy and sensitivity.4. The preliminary analysis of pattern recognition and amino acids metabolism between normal group and the group of lung cancer in human serum as well as the rat liver tissue. The difference analysis of metabolism for amino acids were made through load factor histogram.
