Objective: The changes of NO and substance P concentrations ofoxycodone hydrochloride applications to observe Faure Marin induced painmodel rats before stimulation analgesia and in blood, to investigate the effect ofpreemptive analgesia mechanism of oxycodone hydrochloride.Methods:32healthy adult Wistar rats, male and female, weighing250-300g, were randomly divided into4groups (n=8), normal control group(group A) intraperitoneal injection of0.9%Sodium Chloride Solution2mL;Faure Marin model group (group B) claw palm subcutaneous induced pain onthe right side of rats with microinjector5%Faure Marin100μL injection;oxycodone hydrochloride induced drug group (group C) to pain before, FaureMarin pain induced by intraperitoneal injection of oxycodone hydrochloride30min before0.2mg kg-1; Faure Marin pain induced by intraperitoneal injectionof30min before oxycodone hydrochloride0.2mg kg-1; oxycodonehydrochloride induced pain before and after drug group (D group),30min afterFaure Marin pain induced by intraperitoneal injection of oxycodonehydrochloride0.2mg kg-1. Induced pain after1,10,20,30,45,60,90minobservation of animal pain behavior change, with cumulative pain scoreevaluation of pain behavior.90min with chloral hydrate anesthetized rats after heart, blood3mL, NO and P determination of change of the substanceconcentration.Results:Blood samples of C, NO concentration in the D group comparedwith B group, the difference was statistically significant (P<0.01), between Cgroup and D group, the difference is statistically significant (P<0.05); substanceP concentrations of C, D group compared with B group, the difference wasstatistically significant (P<0.001), between C group and D group, the differencewas statistically significant (P<0.05).Conclusion: There is obvious analgesic effect of oxycodonehydrochloride in Faure Marin rat pain model induced by intraperitonealinjection of30min before and after formalin induced pain, and the analgesiceffect is associated with a lower concentration of NO and P substances in theblood. With the comparison of administration after30min, there is a moresignificant analgesia of administration before30min, which is a morepreemptive analgesia effect.
